摘  要:目的:观察复合培养时,成骨细胞在由快速成型技术构建的具有不同孔隙率的骨支架材料中的增殖情况,考察材料的孔隙率对成骨细胞生长的影响,以探索用快速成型技术(RP)构建骨组织工程支架材料的可行性。 方法:体外分离培养、扩增乳鼠颅骨骨膜成骨细胞,经传代培养作为种子细胞接种于由快速成型技术构建、孔隙率分别为80%,90%,95%的骨支架材料中,复合培养4天及10天后,通过细胞计数及碱性磷酸酶ALP生化染色半定量测定等方法,检测支架材料中细胞增殖情况的变化。结果:1.各孔隙率组的成骨细胞增殖数均呈现随培养时间延长而增长的趋势,细胞随培养时间的增加量则以80%孔隙率组为最低(0.35×105),而90%孔隙率组为最高(2.84×105);2. 各孔隙率组的成骨细胞内碱性磷酸酶的半定量值变化趋势基本与成骨细胞增殖情况一致,均呈现随培养时间延长而ALP相对活性增高的趋势,随培养时间的增加量也仍以80%孔隙率组为最低,90%孔隙率组为最高。结论:快速成型技术构建的骨支架材料的孔隙率对成骨细胞的增殖及ALP相对活性有一定影响,可为组织工程骨支架材料的构形设计提供一定的参考,值得进一步研究。Objective: To study the biocompatibility of bone engineering scaffolds designed and fabricated by CADand rapid prototyping techniques. Methods: Osteoblasts from infant rat calvarial were isolated and expanded in vitro andthen the cells(the 2nd passage) were seeded onto scaffolds with porosity of 80%,90% and 95% at a density of 2.06×106/ml. Cell number and ALP activity were measured after 4 days, 10 days co-culture. Results: (1)The osteoblasts increasedwith culture time for all the three groups, with the lowest increase number of 0.35×106 in the group with porosity of 80%and the highest increase number of 2.84×106 in the group with porosity of 90%; (2) The ALP activity also increased withculture time for the three groups and the differences among groups were shown to be the same as the change of cellproliferation. Conclusions: The studied scaffolds designed and fabricated by CAD and rapid prototyping own goodcellular biocompatibility. The results suggest that the design of scaffolds internal structure like porosity may influence thegrowth of osteoblasts.
