摘 要:目的:探讨白细胞介素-4(Interleukin-4,IL-4)基因多态性与Graves'病(Graves'DiseaseGD)发病易感性的关系。方法:选择汉族GD患者和正常人各100例,采用序列特异性引物聚合酶链反应(PCR-SSP)检测等位基因和基因型。结果:GD组IL-4启动子区-590位置t等位基因频率明显低于对照组(8%对15.5%;P<0.05)。GD组IL-4(-590)c/t杂合子基因型频率显著低于对照组(12%对25%;P<0.05)。结论:在中国汉族人群中,GD患者和正常人之间存在IL-4(-590)等位基因和基因型频率分布的差异。GD患者IL-4启动子区-590位置t等位基因频率下降,可能使IL-4总的转录活性减低,通过免疫炎症反应导致GD发病。Objective:To investigate the association of the polymorphisms of Interleukin-4(IL-4)gene with Graves disease(GD)susceptibility.Methods:One hundred Chinese patients with Graves' disease and one hundred racially matched healthy controls were studied.Alleles and genotypes were determined by polymerase chain reaction and sequence specific primers.Results:The frequency of the variant t allele in the promoter region of IL-4at position-590in GD group was significantly lower than that in control group(8%vs15.5%;P<0.05).The frequency of IL-4(-590)heterozygote genotype in GD group was significantly lower than that in control group(12%vs25%;P<0.05).Conclusion:In Chinese population,there are dif-ferences between GD patients and controls for IL-4(-590)allele and genotype frequencies,the reduced fre-quency of t allele in the promoter region of IL-4at-590position in GD patients may reflect a overall lower transcriptional activity,and this would result in GD through inflammatory immune response.
