摘 要:目的:观察采用ITSF和N2无血清培养方法诱导小鼠胚胎干细胞为神经前体细胞的作用及纤维连接蛋白对神经诱导的影响。方法:小鼠胚胎干细胞的培养和N2无血清条件培养基诱导为神经前体细胞,细菌培养皿法或悬滴法制备胚体,小鼠胚胎干细胞采用NBT/BCIP染色,神经前体细胞的nestin免疫组化染色采用ABC法。结果:细菌培养皿法或悬滴法培养胚体、ITSF或N2无血清培养基均能将小鼠胚胎干细胞诱导为神经前体细胞。纤维连接蛋白预处理培养表面,对胚体的贴壁情况影响不大,但能使无血清培养后存活的神经前体细胞明显增多。结论:小鼠胚胎干细胞在ITSF和N2培养基均能分化为神经前体细胞,在N2培养基中加入纤维连接蛋白能使存活的神经前体细胞数量增多。AIM:To observe the effect of ITSF and N2 serum free medium on the differentiation of neural precursor cells from mouse embryonic stem cells and the influence of fibronectin. METHODS:Culture of mouse embryonic stem cell and its differentiation to neural precursor cells with serum free medium. The embryonic stem cell was stained with NBT/BCIP and the neural precursor cells were stained with immuocytochemistry of nestin. RESULTS:It was found that culturing of embryonic bodies with the bacterial dishes or with hang in drop method, ITSF or N2 serum free medium induced mouse embryonic stem cell to neural precursor cells. The number of survive cells in serum free culturing increased after the surface of dishes was treated with fibronectin, however, the number of embryonic bodies pasted wall had no changes.
