雌激素受体基因多态性与绝经后妇女原发性高血压及血脂关系的研究

摘　　要：目的 :探讨雌激素受体基因多态性在中国汉族绝经后妇女中的分布及其与原发性高血压和血脂水平是否相关。方法 :应用聚合酶链反应和限制性片段长度多态性分析方法 ,检测绝经后妇女中98例原发性高血压患者及90例正常对照者ER基因型 ,并与各组的血脂水平和高血压分级进行比较。结果 :ER等位基因X、x和P、p 频率在原发性高血压组和对照组分别为0 143、0 857、0 444、0 556和0 139、0 861、0 422、0 578。基因型频率分布符合hardy weinberg 平衡定律。XbaⅠ及PvuII酶切多态性中 ,基因型频率、等位基因频率及结合XbaⅠ和PvuII2个酶切多态性分析在组内、组间比较差异均无显著意义 (P>0 05) ,且ER基因型在血脂水平、高血压分级组间比较差异均无显著意义 (P>0 05)。结论 :在绝经后妇女原发性高血压患者中存在ERXbaⅠ和PvuII基因多态性 ,但XbaⅠ和PvuII酶切多态性与原发性高血压无相关性 ,与血脂水平、高血压分级亦无相关性。Objective:To study the relationship between estrogen receptor(ER)gene polymorphism and priˉmary hypertension and serum lipid levels in postmenopausal women.Methods:The ER genes of98patients with primary hypertension and90healthy women who had been over one year after menopause were detected by polymerize chain reactionˉrestriction fragment length polymorphism.Their serum lipid levels were measured siˉmultaneously.Results:In primary hypertension group and control group,the ER allelic frequencies of X,x,P,p and p alleles were0.143,0.587,0.444,0.556and0.139,0.861,0.422,0.587respectively.Genotype distribution and allele frequencies of Pvu II and XbaⅠwere not significant different between the two groups(P>0.05for Pvu II and XbaⅠgenotypes and for Pvu II and XbaⅠallele frequencies).The relationship was not found beˉtween the polymorphisms and the severity of hypertension.Total cholesterol(TC),triglycer(TG),HDLˉC and LDLˉC levels were not significantly different among ER genotype.Conclusion:There is ER Pvu II and XbaⅠgene polymorphism in the postmenopausal patients with primary hypertension,but it is not correlated to primary hypertension and serum lipid levels of the patients.
