奥芬达唑治疗小鼠实验性泡球蚴病效果进一步观察

摘　　要：目的对国产奥芬达唑治疗小鼠实验性泡球蚴病效果观察。方法共进行两批实验 ,按 15 0mg/kg/d× 6 0d口饲给药 ,分别治疗 7只小鼠的泡球蚴病 ,停药后于 30d及 2 4d分别解剖。结果奥芬达唑治愈率 5 7.14 %及85 .71% ,肝与囊平均重量分别为 2 .5 1g及 1.37g。组织切片显示虫囊生发膜中、重度变性、坏死、崩解 ,角质层中等量钙化 ,育囊变性、坏死、钙化。对照药阿苯达唑 (剂量、疗程及解剖时间同上 ) ,治愈率均为 0 ,肝与囊平均重量分别为 3.11g及 1.6 5 g ,切片显示虫囊内可见少许育囊变性、坏死及钙化 ,但亦有囊泡角质层、生发膜完整 ,生发膜细胞增生 ,囊内可见育囊。奥芬达唑对大鼠急性经口毒性试验结果 ,对SD雌鼠的LD50 为 96 87.2mg/kg ,阿苯达唑对SD雌鼠的LD50 为 2 95 0mg/kg。结论奥芬达唑对小鼠实验性泡球蚴病有较好治疗效果 。Objective To study further the therapeutic effect on experimental alveolar echinococcosis and acute oral toxicity of home made oxfendazole. Methods Mice with experimental alveolar echinococcosis were treated with oxfendazole or albendazole in a dose of 150mg/(kg·d)×60 and mice were dissected 24 or 30 days after treatment. The study was carried out for 2 times. In addition, acute oral toxicity of above 2 drugs was studied in white rats. Results In oxfendazole group, the cure rates were 57.14% (4/7) and 85.71% (6/7); average liver and cysts weights 2.51 g and 1.37 g. Pathological section showed that severe degeneration, necrosis and disintegration in germinative membrane, middle size calcification in horny layer, and degeneration, necrosis and calcification in brood capsule could be seen. In albendazole group, the cure rates were 0% (0/7) and 0% (0/8); average liver and cysts weights 3.11 g and 1.65 g. Pathological section showed that in the cysts, a number of brood capsules revealed degeneration, necrosis and calcification, but intact horny layer and germinative membrane as well as cell regeneration, normal brood capsule also existed in the cysts. In SD femele white rats, the acute oral toxicity of oxfendazole was 9 687.2 mg/kg and of albendazole 2 950 mg/kg. Conclusion Oxfendazole showed better therapeutic effect on experimental alveolar echinococcosis and slighter acute oral toxicity than that of albendazole.
