摘  要:目的 研究血清游离三碘甲状腺原氨酸 (FT3)测定的固相酶免疫分析方法。方法 用聚苯乙烯固相载体、亲和纯化的兔抗 T3抗体和 T3-β-半乳糖苷酶标记物建立固相管竞争法、固相板竞争法和固相板顺序饱和法。通过对酶标物进行化学修饰、减少标准品或待测样品的上样量、扩大反应体系和采用顺序饱和法 ,避免或减少甲状腺激素结合蛋白 (TBP)的干扰。结果  3种方法的可测范围均为 0 .4 8~ 4 5 .0 0 ng/ L ,其正常值分别为 (4.34± 1.0 3) ,(4.14± 0 .97) ,(4.0 3± 0 .82 ) ng/ L。正常人与甲状腺功能亢进症患者能明显分开。 3种方法的灵敏度分别为 0 .15 0 ,0 .130 ,0 .0 4 5 ng/ L ,平均批内变异系数分别为 6 .2 % ,6 .4 % ,5 .2 % ,平均批间变异系数分别为13.1% ,12 .4 % ,11.0 %。结论 固相酶免疫分析法基本解决了 TBP的干扰 ,其测定结果的灵敏度和精密度均达到放射免疫分析法 (RIA)Objective To establish different solid phase enzyme immunoassays(EIA) for the quantitation of human serum free triiodothyronine (FT 3). Methods Using rabbit anti T 3 Ab of affinity purification subjected to solid phase support and T 3 β galactosidase conjugate as tracer. We developed coated tube competitive EIA,coated plate competitive EIA and coated plate sequential saturation EIA for quantitation serum FT 3.By the means of minimizing the proportion of sample(and standard)or enlarging reaction system or chemical modification of tracer or sequential saturation. We eliminated or decreased the effect of thyroid hormone binding proteins (TBP).Results The measurement range of the three methods was 0.48~45.00 ng/L. The normal reference interval were (4.34± 1.03 ),(4.14±0.97) and (4.03±0.82)ng/L respectively. There had been significant difference between serum FT 3 level for euthyroid and that for hyperthyroid. The minimal detection limit were 0.150, 0.130 and 0.045 ng/L;intra assay CV were 6.2%, 6.4% and 5.2%, inter assay CV were 13.1%, 12.4% and 11.0%.Conclusions The solid phase serum FT 3 EIA may be concluded that we avoid or decrease the effects of TBP, and are comparable to RIA in respect of sensitivity and precision.
