摘 要:目的研究氢氟酸烧伤中毒后对兔外周血单个核细胞早期凋亡百分率和胞内游离钙浓度([Ca^(++)]i)的影响。方法 用流式细胞仪检测兔烧伤染毒前后外周血单个核细胞的Annexin V变化,以观察其早期凋亡百分率;用Fluo-3/Am荧光探针观察烧伤染毒前后外周血单个核细胞内Ca^(++)平均荧光强度值的变化。结果 12只烧伤中毒兔外周血单个核细胞早期凋亡百分率显著增加,较染毒前比较P<0.01。而12只烧伤中毒兔染毒后1h外周血单个核细胞内[Ca^(++)]i与染毒前比较P>0.05,差异无显著性。结论 本实验提示氯氟酸烧伤中毒引导的细胞凋亡不同于一般认为钙离子浓度升高是大多数细胞凋亡所必需的途径,而是别的凋亡机制。Objective To study poisoning by hydrofluoric acid burn on early apoptosis percentage of single nuclear cell and free calcium concentration in the cell of rabbit. Methods Annexin V changes of single nuclear cell of the. rabbit periphery blood are detected with stream cell detector before and after the rabbit is burned in order to find out the percentage of early apoptosis. And Flou-3/Am fluorescent probe is used to observe the changes of Ca++ average fluorescent strength in the single cell. Results Apoptosis percentage of blood single cell of 12 burned rabbits is obviously increased (P<0.01).No obviously changes of [Ca++]i in single cell of periphery blood (P>0.05), compared before and after poisoning. Conclusion It is indicated cell apoptosis caused by hydrofluoric acid poisoning is different from the general conclusion that rise of calcium ions concentration is the necessary way for most cell apoptosis, but may be other ways.
