摘 要:目的 研究中药复方糖肾宁对早期糖尿病肾病 (DN )大鼠微量白蛋白尿的影响及其作用机制。方法 雄性Wistar大鼠随机分为正常组 (8只 )、造模组 (2 4只 ) ,造模组大鼠采用 3次 (每周 1次 )腹腔内注射链脲佐菌素 (STZ)加福氏完全佐剂(CFA)诱导建立糖尿病模型 ,3周以后测造模组各只大鼠 2 4h尿微量白蛋白 (2 4hU Alb) ,彼此无显著性差异 ,将造模组大鼠分为模型组 (8只 )、蒙诺组 (8只 )、糖肾宁组 (8只 ) ,8周后分别用放免法和比色法测定各组大鼠的肾血液动力学指标和肾组织的超氧化物歧化酶 (SOD)、丙二醛 (MDA)。结果 模型组大鼠血栓素B2 (TXB2 )、TXB2 与 6 酮 前列腺素 (6 keto PGF1α)的比值以及降钙素基因相关肽 (CGRP)显著高于正常组 ,内皮素 (ET)显著低于正常组 (P <0 .0 5) ,肾组织SOD显著下降 ,MDA明显升高 ;糖肾宁组和蒙诺组血浆TXB2 、TXB2 与 6 keto PGF1α 的比值显著低于病理组 ,且糖肾宁组的效果优于蒙诺组 ;与病理组比较 ,糖肾宁组的肾组织SOD含量显著升高 ,而MDA显著下降 (P <0 .0 5)。结论 糖肾宁可降低早期DN大鼠的尿微量白蛋白 ,其作用机制可能与改善糖代谢、调节TXB2 与 6 keto PGF1a的动态平衡并降低血浆CGRP。ObjectiveTo study the modifying effect a nd mechanism of Tangshenning Recipe on micro-albuminuria in rats with early dia betic nephropathy (DN). MethodsMale Wistar rats were r andomly divided into the normal group (n=8)and model group (n=24). Intra peritoneal injecting of streptozotocin (STZ) plus complete Freund’s adjuvant (C FA) was applied once a week for 3 times to induce the DN rats model. Three weeks later,the model group rats were randomly divided into pathologic group (n =8),JP3monopril group (n=8) and Tangshenning Recipe group(n=8)acco rding to the 24 h U-Alb. Each group’sJP renal hemodynamics index and SOD, GS H, MDA in renal tissue were determined by radioimmunoassay (RIA) and colorimetri c method respectively. ResultsThe levels of plasmatic TXB 2, the ratio of TXB 2 and 6-keto-PGF1α, and the CGRP in pathologic grou p were significantly higher than those in normal group. The levels of plasmatic ET decreased obviously, SOD decreased and MDA increased significantly in the rat s’ renal tissue of pathologic group. The levels of plasmatic TXB 2, the ratio of TXB 2 and 6-keto-PGF1α decreased significantly in both Tangshenning Recip e group and monopril group, and the therapeutic effect of Tangshenning Recipe gr oup was better than that of monopril group. SOD was higher and MDA was lower in Tangshenning Recipe group than that in pathologic group. Conclusion The results indicates that Tangshenning Recipe can lower the micro -albuminuria in early DN rats, the mechanism of which probably lies in the modi fication of glycometabolism, the ratio of TXB 2 and 6-keto-PGF1α, the plasma tic CGRP and the renal lipid preoxidation.
