摘 要:研究雌激素受体 (estrogenreceptor ,ER)在成人成骨细胞的分布 ;探讨雌激素调节骨代谢的作用机制及雌激素受体变化在骨质疏松症中的作用。方法 用多次酶消化法进行人成骨细胞体外培养 ;用免疫组织化学法分别检测不同程度的骨质疏松病人和正常对照成人成骨细胞雌激素受体 ;显微镜下观察ER阳性细胞 ;计算各组人群ER阳性细胞率 ;t检验分析各组间ER阳性细胞率的差异。结果 成骨细胞细胞核和 /或细胞质有ER存在 ;各组人群阳性细胞标记率有明显差异。结论 雌激素不仅通过调节钙调节激素间接影响骨重建 ,还可与成骨细胞ER结合调控骨代谢 ;ER存在于成骨细胞的细胞核和细胞质 ;人成骨细胞ER减少可能是绝经后骨质疏松症的主要原因之一。ObjectiveTo examine estrogen receptor (E R) in osteoblasts from adult human and to elucidate the mechanism of estrogen in modulating bone metabolism. MethodsThe cultured osteo blasts were harvested from bone chips by modified sequential digestive enzyme re lease and immunohistochemical assay of ER in osteoblasts were carried out in thr ee groups of female adults: normal control(group 1), patients with moderate oste oporosis(group 2) and patients with serious osteoporosis(group 3). The percentag es of ER-positive osteoblasts from the three groups were compared by t test . ResultsThe brown marks that indicate ER were found i n nuclei and plasma of the osteoblasts, and the percentages of ER-positive oste oblasts among three groups were significantly different. Conclusion ERs exist in nuclei and plasma of the osteoblasts. Estrogen may mod ulate bone metabolism through binding ER in nuclei and plasma of the osteoblasts . The reduction of ER of osteoblasts may play an important role in the pathogene sis of postmenopausal osteoporosis.
