摘  要:分析了部分皂化的二(2-乙基己基)磷酸酯/正庚烷反相微乳液萃取Zn2+/胰蛋白酶、Ni2+/胰蛋白酶前后体系的紫外光谱和透射电镜。结果表明,在胰蛋白酶萃取有机相中,蛋白质、两亲分子及过渡金属离子3者之间存在着复杂相互作用,金属离子的种类、浓度、水相的pH值对胰蛋白酶的羰基和芳香族残基的微环境均有不同程度的影响,并导致胰蛋白酶萃取率的变化。同时,从萃取前后微乳液和胰蛋白酶的形貌可见,胰蛋白酶可较好的被萃取进入反相微乳液的水核中,过渡金属离子的加入则使微乳液发生破乳现象。Di(2ethylhexyl)phosphoric acid ester/heptane systems were saponified by NaOH solution to form W/O microemulsions. Zn2+/trypsin and Ni2+/trypsin were extracted by the microemulsions. The extractive organic phases were studied by ultraviolet spectra and TEM. The results showed that there were complicated interactions among the trypsin, surfuctant and metal ions in the extractive organic phases.The kind of ions and ionic strength and pH value of aqueous solution affected the microenvironment and the solubility of trypsin in the organic phases. By comparing the TEM photos of microemulsion with that of the organic phases,it was seen that trypsin was successfully extracted in the water core of microemulsion and the microemulsion was destroyed by adding metal ions.
