摘 要:利用Ashby培养基从不同林龄的降香黄檀根际土壤中分离高效固氮菌,为专用生物复合肥的研制提供理论基础。固氮酶活在75.30~432.76 nmol h-1 ml-1之间的菌株有21株,其中菌株JT-N305的固氮酶活最高。其中15株菌株具有分泌IAA的能力,其分泌量在9.76~41.21μg ml-1。结合形态学、生理生化特性及16S r DNA序列分析,初步鉴定JT-N301属于Burkholderia anthina、JT-N304属于Enterobacter ludwigii、JT-N305属于Burkholderia seminalis。菌群的研究表明:3株菌株混菌培养,其固氮酶活高达498.66 nmol h-1 ml-1;相对于其它氮源而言,菌群更易于吸收铵盐;而菌群的最佳碳源为葡萄糖。In order to provide th eoretical basis for the development of the special biological compound fertilizer, the high- efficiency Azotobacter were screened from different ages of Dalbergia Odorifera rhizosphere soil by Ashby medium. Of the isolate strains, 21 strains possessed higher nitrogen- fixing capacity, with the range of nitrogen live from 75.30 nmol h- 1m L- 1to 432.76 nmol h- 1m L- 1, among which strain JT- N305 showed the highest nitrogen- fixing capacity. 15 of the 21 strains had the ability to produce IAA, with the range of IAA- producing from 9.76 μg m L- 1to41.21 μg m L- 1. Based on the morphology, physiological and biochemical property and 16 S r DNA sequence analysis,JT- N301, JT- N304 and JT- N305 were preliminarily identified as Burkholderia anthina, Enterobacter ludwigii and Burkholderia seminalis, respectively. The research of 3 strains mixed culture showed that the nitrogenase activity was up to 498.66 nmol h- 1 m L- 1. Compared with other nitrogen sources, ammonium salt was more likely to be absorbed by the mixed microflora. The optimum carbon source was glucose.
