风湿性心脏病二尖瓣狭窄患者外周血淋巴细胞TGF-β_1 mRNA的表达

摘　　要：目的 :探讨外周血淋巴细胞 TGF- β1m RNA表达与风心病二尖瓣狭窄之间的关系。方法 :用逆转录聚合酶链反应 ( RT- PCR)半定量技术 ,对 1 5例风心病二尖瓣狭窄患者与 1 5例正常人 TGF-β1m RNA的表达进行检测。以 TGF-β1/β- actin光密度值之比作为半定量结果。结果 :两组受检者均可见 TGF-β1m RNA的表达 ,风心病二尖瓣狭窄患者 TGF-β1m RNA的表达明显高于正常人 ( t=2 .1 34,P<0 .0 5 )。结论 :风心病患者不仅存在瓣膜局部 TGF- β1的表达增强 ,还存在外周血淋巴细胞 TGF- β1的表达增强 。Objective:To study the relation between rheumatic heart disease and the expression of TGFβ 1 mRNA in peripheral blood lymphocytes.Methods:RT PCR was used to detect the expression of TGF β 1 mRNA in 15 patients with rheumatic disease complicated by mitral stenosis and in 15 healthy cases.Results:The expression of TGF β 1 mRNA and β actin could be detected in two teams.TGF β 1 mRNA level in patients of mitral stenosis with rheumatic heart disease was significantly higher than that in healthy ones(0.698±0.125 vs 0.326±0.108 , P <0.05).Conclusion:The expression of TGF β 1 mRNA is increased in lymphocytes in patients of mitral stenosis with rheumatic heat disease.The mechanism of increased TGF β 1 mRNA expression needs further investigation. [
