摘  要:目的探讨柯萨奇病毒B3感染与心肌细胞凋亡的关系。方法将 10 0TCID50 柯萨奇病毒B3腹腔注射BALB/c小鼠 ,于接种后 3、5、7、9和 14天各处死 5只 ,取心脏 ,将右半心脏匀浆 ,冻融 3次后离心取上清 ,用HeLa细胞分离病毒并滴定其效价 ;左半心脏进行组织病理学检查和用TUNEL方法检测凋亡细胞。结果接种病毒后第 3天和第 5天光镜观察可见心肌纤维崩解、断裂 ;第 9天和 14天间质纤维增生 ,填充坏死心肌吸收后的间隙。接种后 3~ 9天心肌内可分离到病毒 ,第 7天病毒效价达高峰 ,第 9天下降 ,第 14天分离不到病毒 ;接种后 3~ 14天在心肌内检测到凋亡细胞 ,凋亡细胞数量呈逐渐增加趋势。结论柯萨奇病毒B3型感染BALB/c小鼠可以引发心肌细胞凋亡 。ObjectiveTo investigate the relationship between Coxsackievirus B3 induced murine myocarditis and apoptosis of myocardium. MethodsBALB/c mice were inoculated intraperitoneally with 100 TCID 50 Coxsackievirus B3(CVB3). On the 3rd, 5th, 7th, 9th and 14th day post inoculation, respectively five mice were killed and the hearts were removed aseptically. The right parts of the mice hearts were homogenized, frozen and thawed three times, and cleared of cellular debris by low speed centrifugation. The viruses in supernatants were titrated on HeLa cell monolayers.The histopathology and cardiomyocyte apoptosis in the sections of the left parts of the hearts were studied. ResultsThe sections of heart tissue showed degeneration of the cardiac myocytes from the 3rd to 5th day, then followed by progressive disintegration of cardiac muscle fibers on the 7th day. On the 9th and 14th day interstitial fibrosis and cellular infiltration were observed. The virus could be separated from the hearts from the 3rd to 9th day, and the titer reached the peak on the 7th day. Apoptosis was widely spread in both cardiac myocytes and interstitial cells on myocardial sections from the 3rd to 14th day. The number of apoptosis cells showed a tendency of ascending until the 14th day. ConclusionCardiomyocyte apoptosis played a critical role in Coxsackievirus B3 induced murine myocarditis.
