海风藤对局灶性脑缺血治疗作用的实验研究

摘　　要：目的 :探讨海风藤提取物对老龄大鼠局灶性脑缺血的治疗作用。方法 :建立光化学诱导的老龄大鼠局灶性脑缺血模型 ,利用TTC染色、HE染色、TUNEL标记、原位分子杂交观察海风藤提取物对缺血灶大小、血脑屏障、缺血灶周围坏死细胞、凋亡细胞及p5 3基因表达变化的影响。结果 :海风藤治疗组大鼠局灶性脑缺血后血脑屏障破坏明显减轻 ,缺血灶周围坏死细胞、凋亡细胞及P5 3阳性细胞的数量较未干预组显著减少 ,梗死灶直径也有减少的趋势。结论 :海风藤具有抵抗缺血后神经细胞DNA损伤、减轻迟发性神经元死亡及神经细胞坏死、发挥缺血性脑保护作用。Aim:To investigate the effective of Piper kadsura Ohwi at the focal cerebral ischemia in aged rats.Methods:The model of focal cerebral ischemia induced by photochemistry in aged rat was established and using TTC,HE,TUNEL and in situ hybridization techniques, we observed the effects of Piper kadsura Ohwi on the size of ischemia,blood-brain barrier,apoptosis and p53 gene expression in perifocal area.Results:The destruction of blood-brain barrier was relieved and the number of necrotic,apoptotic and P53 positive cells were reduced significantly in penumbra area after injection of Piper kadsure Ohwi in focal cerebral ischemia.Conclusion:Piper kadsura Ohwi may play protective effect by anti-DNA damage and by reducing delayed neuronal cell death and necrosis after focal cerebral ischemia in aged rats.
