重庆地区乙型肝炎病毒B/adw2和C/adrq+亚型PreS/S基因参照序列的初步建立

摘　　要：目的　建立重庆地区不同亚型乙型肝炎病毒 (HBV)流行株PreS S基因参照序列。方法　扩增 18例HBV慢性无症状携带者的PreS S基因 ,应用同源性分析和对齐比较等方法确定基因型 血清型 ,并选同HBV亚型的PreS S拟定参照序列。结果　 9株基因型B 血清型adw2、6株基因型C 血清型adrq +、3株基因型B 血清型ayw 1;建立的重庆地区基因型B 血清型adw 2、基因型C 血清型adrq +HBV流行株PreS S参照序列 ,与华东华南、东北华南地区同亚型的参照序列比较分别有6个、13个核苷酸差异 ,均可引起 4个氨基酸差异。结论　初步建立了重庆地区HBV基因型B 血清型adw2、基因型C 血清型adrq +HBV流行株PreSObjective To develop consensus sequence o f HBV PreS/S with different subtype in Chongqing of China. Methods The gene of PreS/S of HBV in 18 AsC was sequenced .The genotype and serotype of HBV were determined. The main HBV strain prevailin g in Chongqing was identified to establish its consensus sequence. Results It was found that 9 strains were genotype B/serot ype adw2,6 were genotype C/serotype adrq+ and 3 were genotype B/serotype ayw1. T he consensus sequence of PreS/S of HBV with genotype B/serotype adw2 and genotyp e C/serotype adrq+ were established. There were 6 nucleotide variants which caus ing 4 amino acids change between consensus sequence of PreS/S with genotype B /serotype adw2 in Chongqing and in Southeast of China (homology showed 99.5 % and 99.0 % respectively).There were 13 different sites that caused 4 ami no acids change between consensus sequence of PreS/S with genotype C/serotype ad rq+ in Chongqing and in Northeast and South of China (homology showed 98.9 % and 99.0 % respectively). Comparing the two consensus sequence of PreS/S gene of different subtype HBV in Chongqing,there was 95 variants which caused 40 ami no acids variants (the homology was 92.1 % and 90.0 % respectively). Conclusion The consensus sequence of PreS/S of hepatitis B virus with genotype B/serotype adw2 and genotype C/serotype adrq+ prevailing in Chongqing was established.
