高效液相色谱测定经PT-系列氧化铝小柱预处理后狗血清中儿茶酚胺

摘　　要：氧化铝吸附法提取生物样本中的儿茶酚胺已被广泛地应用,但在操作上需反复洗涤、离心,比较麻烦,本文探讨了用国产PT-系列酸性氧化铝小柱进行预处理的条件,用内标法测定了肺动脉高压情况下的狗血清中去甲肾上腺素(NE)和肾上腺素(E)含量的变化,得到较为满意的结果。A high performance liquid chromatographic method with electrochemical detection for measuringnoradrenaline(NE) and adrenaline (E ) in the serum of dog is presented. NE and E were extracted with PT series Al2O3 cartridge, and chromatography was performed on a reversed phase column using DL-camphorsulfonic acid as ion pair reagent. The recoveries of NE and E were 57. 33±1. 15 and 53. 67 ± 4.72%, respectively. The recovery curves were linear within the range of 1-30 ng/ml for NE and 2-60 ng/ml for E. The results showed that the elevation of NE in serum of dog which suffered from high intrapulmonic pressure was significant in comparison with the controls.
