新疆哈萨克族人血管紧张素原基因T174M多态性与原发性高血压的相关性研究

摘　　要：为探讨血管紧张素原(AGT)基因的T174M多态性与新疆哈萨克族人原发性高血压的关系,以人群为基础进行病例-对照研究,应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析技术,检测128例原发性高血压患者与74例正常对照者的AGT的T174M的多态性。结果显示新疆哈萨克族人原发性高血压AGT基因型频率为TT:78.91％,TM:20.31％,MM:0.78％;T174与M174等位基因频率分别89.06％、10.94％。而对照组其相应频率分别为78.38％、21.62％、0％、89.19％、10.81％,两组相比无显著性差异(P>0.05)。按性别分组后,男女等位基因频率两组相比亦无显著性差异(P>0.05)。提示新疆哈萨克族人的AGT基因T174M多态性和新疆哈萨克族高血压无关的结论。To investigate the relationship between the T174M allele polymorphism of angiotensinogen (AGT) gene and the essential hypertension in Kazakans of Xinjiang, Polymerase chain reaction and restriction fragment length polymorphism (PCRRFEP) methods were used to detect the T174M polymorphism of AGT gene in Kazakans including 128 patients and 74 normotensive control. The genotype frequencies of T174M were TT 78.91%; TM 20.31%, MM 0.78% respectively; T174 and M174 allele frequencies were 89.06%, 10.94% respectively in hypertension group. While the corresponding frequencies were 78.38%, 21.62%, 0%; 89.19%, 10.81% respectively in control group. There are no significant difference in MM174 genotype and M174 allele frequencies of angiotensinogen between hypertension and control groups(P>0.05). In addition, T174 and M174 allele frequencies had also no statistic significance in the subgroups classified according to sex(P>0.05). It is concluded that the M174 variant of the angiotensinogen may not play a critical role in the pathogenesis of hypertension in Kazakans of Xinjiang.
