摘 要:取河北省东部沿海及内地不同生态条件下生长的14个基因型的野生大豆为试验材料,在含不同质量分数NaCl(0、0 2%、0 4%、0 6%、0 8%、1 0%)的继代培养基上观察愈伤组织的耐盐性,并进行苗期耐盐性鉴定。结果表明:不同基因型材料之间愈伤组织的耐盐性有明显差异,耐盐性强的可在0 8%NaCl条件下生存,低的只能忍耐0 2%NaCl含量;试验确定野生大豆愈伤组织耐盐性鉴定适宜的NaCl含量为0 6%;盐碱条件下生长的野生大豆愈伤组织一般耐盐性较强,非盐碱环境下生长的野生大豆也存在着愈伤组织耐盐性较强的类型,试验发现细胞水平耐盐性强的材料在苗期也表现较强的耐盐性。The 14 genotypes of wild soybean growing at different condition in eastern of Hebei province were studied in this experiment. Hypocotyls and cotyledon were used as explants, callus was induced on medium containing different composition. The salt-tolerance of callus was observed at different medium containing 0,02%,04%, 06%,08%,10% NaCl. Ability of salt-tolerance was identified through growthamount,quality,becoming brown and death ratio of callus. The results showed that MS+20 mg/L 2,4-D+05 mg/L 6-BA+sucrose 5% +agar 08% is the best medium for callus induction. Ability of salt-tolerance of callus had obvious difference among the different genotypes. The stronger salt-tolerance genotypes could survive at the concentration of 08% NaCl. The ability of salt-tolerance of 6,7,10,11,12,14 was stronger, the growth amount and quality of callus descended with the rising of concentration of NaCl. The experiment indicated that identity indicator of salt-tolerance of wild soybean is 06% NaCl. Generally, the wild soybean from saline soil have stronger ability of salt-tolerance. The results suggested that the relation of salt-tolerance ability at cell level and plant level should study further.
