摘 要:目的 比较正常妊娠与自然流产模型母胎界面细胞因子表达特征的差异 ,探讨T辅助细胞 (Th) 1型和 2型 (Th1 /Th2 )细胞因子在介导流产发病中的作用机制。 方法 建立正常妊娠模型CBA×BALB/c和自然流产模型CBA×DBA/ 2。采用酶联免疫吸附试验 (ELISA)测定两组模型孕第 9和 1 4天母胎界面组织培养上清中Th1 /Th2型及相关细胞因子表达水平 ;以白细胞介素 4/干扰素 γ(IL 4/IFN γ)比值作为衡量Th2 /Th1型免疫调节平衡的指标。 结果 孕第 9天 ,与正常妊娠模型相比 ,自然流产模型母胎界面IL 4、IL 1 0和转化生长因子 (TGF) β2的表达显著降低 (P <0 .0 5) ;IFN γ、肿瘤坏死因子 (TNF) α的表达显著升高 (P <0 .0 5) ,IL 4/IFN γ比值显著降低 (P <0 .0 5)。至孕第 1 4天 ,自然流产模型母胎界面IFN γ、TNF α表达仍显著高于正常妊娠模型 (P <0 .0 5) ,IL 1 0表达、IL 4/IFN γ比值仍低于正常妊娠模型 (P <0 .0 5) ,IL 4和TGF β2在两组模型中的表达无显著差异。两组模型孕第 9和 1 4天母胎界面均未测到IL 2的表达。 结论 与流产模型相比 ,无论在妊娠早期或中晚期 ,正常妊娠模型母胎界面局部均呈现明显的Th2型免疫偏倚。母胎界面Th1Objective: Comparing the characters of cytokine expression at maternal-fetal interface in normal pregnancy and in spontaneous abortion mouse models to probe the roles of Th1 and Th2 cytokines in spontaneous abortion. Methods: The abortion-prone CBA×DBA/2 matings were established as the model of spontaneous abortion and the nonabortion-prone CBA×BALB/c matings were used as the model of normal pregnancy. The ectoplacental cell cones and decidua were cultured for 48 hours at day 9 of gestation, and the placenta and decidua were cultured for 24 hours at day 14 of gestation respectively, the supernatants were then collected and the levels of Th type-1 and type-2 cytokines were determinated by ELISA. The ratio of IL-4/IFN-γ was used as the index to evaluate the immune regulation balance between Th1 and Th2. Results: The expressions of IL-4, IL-10 and TGF-β2 at the maternal-fetal interface of CBA×DBA/2 matings were significantly reduced(P<0.05), while the expressions of IFN-γ and TNF-α were significantly increased(P<0.05), and the ratio of IL-4/ IFN-γ reduced as compared with that of CBA×BALB/c matings. At day 14 of gestation, the expressions of IFN-γ and TNF-α in CBA×DBA/2 matings were still higher and expression of IL-10 was still less than that of CBA×BALB/c matings, the ratio of IL-4/IFN-γ reduced significantly(P<0.05). There was no significant difference in IL-4 and TGF-β2 expressions between the two models Conclusion: Whenever at the middle or the late stage of gestation, it is obvious that a predominant Th2 bias exists at the maternal-fetal interface of the CBA×BALB/c matings as compared with that of CBA×DBA/2 matings. Abortion is the inevitable result because of the regulation disordor in the balance between Th1 and Th2 cytokines.
