摘 要:目的 观察蛋白C在鼠糖尿病模型中的改变及通心络胶囊干预的影响。方法 将SD大鼠随机分为两组 ,一组经链尿佐菌素诱发的糖尿病模型 (DM)组 ,另一组为血糖正常对照组 (C) ,两组均再随机分为DM1组、DM2组、C1组、C2组 ,其中DM2、C2组给予通心络治疗 5周。治疗前后称体重、监测血糖 ,采用发色底物法测定血浆蛋白C活性。结果 DM1、DM2、C1和C2组血浆蛋白C活性分别为 4 95± 1 2 4% ,6 48± 2 0 2 % ,3 61± 1 12 %和 5 0 8± 1 43 %。DM2组高于DM1组 ,DM1组高于C1组 ,C2组高于C1组 ,差别有显著性意义 (p <0 0 5 )。结论 蛋白C活性在鼠糖尿病早期就出现增强 ,蛋白C参与糖尿病性心脑血管病变的发生和发展过程。通心络能使正常鼠蛋白C活性增强 ,也能使蛋白C活性本已增强的糖尿病鼠的蛋白C活性进一步增强 ,可能对糖尿病致心脑血管疾病有一定的治疗和预防作用。Objective To investigate the change of the activated protein C in diabetic rats and the effect of Tongxinluo capsule. Methods SD rats were randomly divided into 2 groups: streptozotocin-induced diabetic model (22 cases, DM group), and control group (20 cases, C group). DM group and C group were respectively and randomly divided into 2 subgroups: DM1, DM2, C1 and C2. The subgroup of DM2 and C2 were administrated with Tongxinluo capsule for 5 weeks whereas DM1 and C1 were given normal saline. Body weight and blood sugar level of all rats were measured respectively before and after treatment, and the levels of activated of protein C were measured. Results The levels of activated protein C in DM1, DM2, C1 and C2 group were 4.95±1.24%,6.48±2.02%, 3.61±1.12% and 5.08±1.43% respectively. DM2 group had a higher level of activated protein C than that in DM1 group, and the same situation existed as DM1 group was compared with C1 group, and C2 group was compared with C1 group, all with statistically significant differences. Conclusions The levels of activated protein C upregulates in early diabetic rats, which may play a role in the occurrence and development of diabetic cardiovascular and cerebrovascular diseases. Tongxinluo capsule can increase the activity of protein C in normal rats and diabetic rats in which the activity of protein C has increased, which may contribute to the prevention and treatment of cardiovascular and cerebrovascular diseases.
