摘　　要：目的　观察造血抑制因子TNF α对人脐血基质细胞体外扩增的效果。方法　采用Dexter培养法 ,加入含TNF α的不同细胞因子组合 ,计数培养 2 1d时的基质细胞集落数 ,测定培养 7,1 4 ,2 1 ,2 8d的细胞周期。结果　对人脐血基质细胞的扩增效果 ,TNF α单用与对照组比较 ,P <0 .0 1 ,TNF α+SCF +BFGF与其他实验组比较 ,P <0 .0 1 ;人脐血基质细胞扩增培养不同时相点的G2 +M +S期细胞分别占 :2 5 .6 % ,2 9.8% ,36 % ,2 2 .1 %。结论　TNF α单独或联合使用对人脐血基质细胞有较明显的扩增作用 。Objective To study the effects of TNF α on human umbilical cord blood stromal cell(HUCBSC) colony. Methods Using the methods of ex vivo culture of cord stromal cells,and adding the TNF α and different cytokine groups. At the 21d in culture, the number of HUCBSC colony was counted and the cell cycle was detected in 7,14,21,28d respectively.Results The group2 which added TNF α had a significant difference compared with the control groups,( P <0.01).Furthermore, the group of SCF+bFGF+TNF α had a marked diversity when compared with other groups,( P <0.01) ;the ratio of HUCBSC in G 2+M+S phase was 25.6%, 29.8%,36%and22.1% individually.Conclusion The TNF α has promoting effects on the expansion of HUCBSC in vivo. The effect is better when it associate with hematopoitic growth factor .
