摘 要:目的 探讨三羟异黄酮抑制红系祖细胞分化的机制。方法 选择三羟异黄酮处理脐血CD34+ 细胞 ,采用红系爆式集落计数、免疫表型分析等实验方法 ,观察三羟异黄酮对红系祖细胞分化的抑制作用 ,同时用Westernblot分析检测EphB4酪氨酸磷酸化变化。结果 1 0~ 90 μmol/L三羟异黄酮作用 7~ 1 4d可抑制红系爆式集落形成和 glycophorinA蛋白表达 ,同时红系祖细胞EphB4酪氨酸磷酸化水平降低 ,并且三羟异黄酮抑制红系祖细胞分化与剂量呈正相关。Objective To investigate the molecular mechanism of inhibiting differentiation on erythroid progenitor cell by genistein. Methods Human umbilical cord blood CD34 + cells were dealed with genistein.The index to reflect differentiative inhibition of erythroid progenitor cells was determined with BFU E and erythroid cell surface marker test.Western blot was used to analyse the level of tyrosine phosphorylation of EphB4.Results The forming of BFU E and expression of glycophorin A protein were inhibited after they were given (10 90)μmol/L genistein 7 14 days. Tyrosine phosphorylation level of EphB4 was decreased at the same time.There was positive relation between the dose of genistein and differentiative inhibition of erythroid progenitor cells. Conclusion The change of tyrosine phosphorylation of EphB4 might be the molecular mechanism of inhibiting differentiation of erythroid progenitor cells.
