Selection of New Germplasm Resource Using the Technique of Pollen Tube Pathway Transfer Exogenous DNA into Recipients of Oats

摘  要:采用花粉管途径将不同倍性燕麦以及花生、大豆、小麦等不同作物外源DIA导入裸燕麦早熟品种品二号、中晚熟品种冀张莜四号和晚熟品种冀张莜五号,获得了较为广泛的变异,经过逐代选择鉴定,育成了一批裸燕麦新种质。本研究以GUS报告基因为外源目的基因用同样方法导入燕麦,从其导入一代(GT)中检测出了表达GUS活性的阳性植株。Using a series of naked oat variety as recipients, such as early-ripe PIN2 mid-late maturing Jizhangyou 4 and late-maturing Jizhangyou 5 and varied-ploidy oats, peanut, soybean and wheat, etc. as the donor, Exogenous DNA was transfered into recipients by pollen tube method, and variations had been obtained. After selected and identified generation-by-generation a series of germplasm resource of naked oats were developed . In order to further test the efficiency of transformation by pollen tube method GUS report gene was introduced as exogenous target gene into oats. In the colony of GT1 ,a great number of positive plants were inspected what expressed the activity of GUS gene.
