(1,3)-β-D-葡聚糖ELISA检测方法建立及初步临床应用

摘　　要：目的 建立一种检测各种生物体液中(1,3)-β-D-葡聚糖的ELISA法。方法 以半乳糖神经酰胺为包被物,鼠抗(1,3)-β-D-葡聚糖单克隆抗体、生物素化抗鼠IgG和辣根过氧化物酶标记亲和素为检测系统建立ELISA法。结果 该法(Y)在葡聚糖含量低于800 ng/L时线性良好,并具有良好的重复性(CV:3.97％-6.9％)和满意的回收率(100.8％);与显色鲎试验(X)有良好的相关性。Y=1.004X-0.915,r=0.978 3。结论 该法操作简便、结果可靠,适用于临床普通实验室。Objective To establish an enzyme-linked immunosorbent assay for (1, 3)-β-D-glucan in all kinds of biological fluid. Methods Precoating microplate with galactosylcermide,detecting system includes mouse anti-(l ,3)-β-D-glucan IgG and HRP-ABC compounds. Results This assay had a wide linear range (0-800 ng/L) and good reproducibility (CV: 3. 97% -6. 9%), the average recovery rate was 100. 08%. Compared with chromogenic LAL,the relative coefficient (r) was 0. 978 3,y(ELISA) = 1.004X -0. 915. Conclusion This ELISA is a simple,reliable assay ford ,3)-β-D-glucan,and can use in general clinical laboratories.
