摘 要:目的以亚理想环境下培养的心肌细胞为模型 ,分析小牛血去蛋白提取物 (DECB)对心肌细胞生长方面的作用。方法对结晶紫和 [3H] TdR摄取量的测定反映心肌细胞的增生程度 ;用Lowry′s法测培养心肌细胞的蛋白质含量 ;通过对D [6 3H]葡萄糖测定反应心肌细胞葡萄糖摄取量 ;通过镜下计数法测定培养心肌细胞搏动频率。结果DECB能够促进细胞增生 ;增加细胞蛋白质含量 ;能够增加葡萄糖的吸收和增加细胞搏动频率。结论在亚理想培养条件下 。PurposeTo study the effects of deproteinated extract of calf blood(DECB) and the possible function mechanism on the cell proliferation, protein content, glucose transport and contracting frequency under sub-optimal culture conditions.ie.0.4 % calf serum.MethodsCell proliferation was determined by the crystal violet staining uptake and [ 3H]-TdR uptake. Protein content was assayed wit h Lowry′s method. Glucose transport was measured by D-[6- 3H]Glucose uptak e. Contracting frequency of cultured myocardial cells was counted under the inve rted microscope.ResultsDECB may be beneficial to increas ing in culture cell density, protein content, glucose transport, and contracting frequency under sub-optimal culture conditions,i.e.0.4% calf serum.Con clusionDECB can promote the myocardial cells proliferation in vitro under sub-optimal culture conditions,i.e.0.4% calf serum.
