摘  要:目的 :建立非衍生化 气相色谱法 (GC)测定人血清游离脂肪酸 (FFA)谱的方法。 方法 :采用GC法测定血清FFA。色谱柱为化学键合相FFAP毛细管柱 (30m× 0 .32mmI.D ,涂层厚 0 .2 5 μm) ,火焰离子化检测器。血清样品前处理方法为 :以庚酸 (C7∶0 )和正十七酸 (C1 7∶0 )为内标 ,用正己烷 异丙醇 (4∶1 ,v/v)提取血清中FFA后直接进样。 结果 :除乙酸和丙酸外 ,其余脂肪酸的回收率在 81 .0 8%~ 1 1 8.2 9%。日内变异系数均 <8.95 % ,日间变异系数均 <8.4 7% ,用各FFA与内标色谱峰面积的比值对浓度做标准曲线 ,γ2 在 0 .992 5~ 0 .9998。 结论 :该方法简便可靠 ,符合生物样品的分析要求 。Objectives: A method of underivatized gas chromatography (GC) was established for the determination of free fatty acid (FFA) profile in human serum. Methods: A capillary column with a chemically bonded free fatty acid phase (FFAP) was used to separate free fatty acid profiles with a programmed rising temperature system. Free fatty acid profile were detected by flame ionization detection (FID). n Heptanoic acid(C7 ∶ 0)and Heptadecanoic acid (C17 ∶ 0) were used as internal standard (IS). The free fatty acids were extracted from serum by hexane isopropanol (4 ∶ 1, v/v ). Results:Except C2 and C3, the recoveries ranged from 81.08 % to 118.29% .The intra and inter day coefficient of variation were less than 8.95% and 8.47% respectively. Conclusions:This method is simple, reliable and suitable for the determination of FFA in biological sample.
