摘  要:目的 研究临床分离的结核分枝杆菌菌株感染人巨噬细胞所引起的表达变化 ,寻找与其它细菌在诱导细胞表达方面的共性和特性。方法 用 192 0 0个基因的寡核苷酸芯片比较临床分离细菌感染U937前后细胞基因的表达变化 ,以生物信息学统计分析其它细菌引起宿主细胞的基因表达变化。结果 结核分枝杆菌感染导致 1395个基因 (7.2 6 % )差异表达 ,特别显著的是细胞因子、锌指蛋白、转录因子和凋亡有关因子。与沙门菌、铜绿假单孢菌、金黄色葡萄球菌、单孢李斯特菌、百日咳菌等细菌感染引起的免疫反应具有相似之处。结论 结核分枝杆菌感染与其它细菌感染引起的免疫反应在转录水平具有共性 ,也富有特性。锌指蛋白和fibronectin受体等因子在抗结核分枝杆菌感染免疫中的作用值得深入研究。Objective To investigate the immunity elicited by the clinical isolates of Mycobacterium tuberculosis , and to find the common theme and distincts in pathogen invasion. Methods Oligonucleotide microarray containing 19 200 genes were used to compare the gene expression profiling of macrophage U937 before and after exposure to the strain, bioinformatics comparison of the published data of other pathogen that caused host cell differential expressed gene were employed. Results Among the 19?200 genes monitored, 1?395 (7.26%) genes were differential expressed. Most prominent ones are the cytokines, zinc finger proteins, transcriptional factor, and apoptosis-related factor. The differential expressed genes shared the same pattern of those elicited by the Salmonella, Pseudomonas aureginosa, Staphylococcus aureus, Listeria monocytogenes, Bordetella pertussis . Conclusion The antimycobacterial immunity shared the pattern of other bacterial pathogen with its unique features. Zinc finger protein, fibronectin receptor are of potential candidate of drug target.
