摘 要:目的 分离、纯化再生障碍性贫血 (简称AA)患者骨髓CD3 4+ 细胞 ,探讨其发病机制。方法 对4 2例AA患者 ,以常规方法进行骨髓液采集和有核细胞分离 ,用单克隆抗体 免疫磁珠分离系统 (MACS)分离、纯化骨髓CD3 4+ 细胞 ,用碱性磷酸酶 抗碱性磷酸酶 (APAAP)法作CD3 4+ 细胞的纯度检测。结果 骨髓CD3 4+ 细胞纯度达 95 %~ 99% ,AA患者骨髓CD3 4+ 细胞多呈单个、散在分布 ,着色较淡 ;而对照组骨髓CD3 4+ 细胞多呈均匀或丛簇状分布 ,着色较深。结论 应用MACS分离AA骨髓CD3 4+ 细胞 ,特异性强、纯度高 ,是研究AA骨髓CD3 4+ 细胞功能、形态的最直接、客观。Objective To separate and purify bone marrow CD 34 + cells from patients with aplastic anemia and to explore the pathogenesis of aplastic anemia.Methods Bone marrow of 42 patients with aplastic anemia was accumulated and eucells were separated.Then bone marrow CD 34 + cells were separated and purificated with monoclone antibody immunomagnetic beads system(MACS),before purity of acquired CD 34 + cells was detected with alkaline phosphatase antialkaline phosphatase (APAAP).Results The purity of acquired bone marrow CD 34 + cells got somewherer on 95% to 99% and CD 34 + cells of aplastic anemia patients stained with light colour were individually distributed,while CD 34 + cells of the control group evenly distributed or in clumps,heavily stained.Conclusion Separating bone marrow CD 34 +cells of aplastic anemia with MACS,owing to its high pecularity and purity,is the most direct,objective and accurate method to study the form and function of bone marrow CD 34 + cells so far.
