鸽新城疫病毒的分离、鉴定和形态观察

摘  要:从以神经症状为主,有很高传染性和死亡率的病鸽中分离到1株病毒,分离株对鸽有致病性,病死鸽的脑、肝组织悬液及毒株的鸡胚传代材料能凝集鸡红血球,用禽Ⅰ型副粘病毒抗血清,由血凝抑制试验得知该病毒是禽Ⅰ型副粘病毒,用差速离心方法处理增殖病毒的鸡胚尿囊液,可得纯度较高的病毒提纯物,负染后,由电镜观察可见病毒呈多形态,一般为圆形,直径约100~200nm,有“鲱鱼骨”状核衣壳,外被约10nm厚的囊膜,上有整齐排列的8~9nm的纤突。A virus was isolated from pigeons showing nervous disoders with high infectivity and high mortality. The isolates proved to be pathogenic for pigeon. The brain and liver suspensions from sick pigeon or embryonic fluids from eggs inoculated with brain and liver suspensions agglutinated chicken erythrocytes. This virus was confirmed as Avian Paramyxovirus type Ⅰ in haemagglutination inhibition test using antiserum against Avian Paramyxovirus type Ⅰ. Using a method of differential centrifugation to treat the virus-infecsted allantoic fluid, we can get highly purified virus. After negative staining, we can see that the viruses are very pleomorphic particles, generally they are rounded 100~200nm in diameter and have 'herring bone' nucleocapsid covered with envelope about 10nm in length, there are 8~9nm projections on the surface of envelopes in most electron micrographs.
