摘 要:用体外骨髓贴壁细胞长期传代法,建立一株骨髓基质细胞QXMSC1,经鉴定可能为巨噬细胞来源。用5Gyγ射线照射BALB/c小鼠建立造血功能缺损动物模型,将QXMSC1细胞输入到照射小鼠体内,用半固体琼脂法测定粒-巨噬系祖细胞,甲基纤维素法测定红系祖细胞,血细胞自动计数仪测定外周血各项血液学指标。结果表明,在照射后第10天,骨髓基质细胞可增加辐射损伤小鼠骨髓有核细胞数,增加CFU-GM,CFU-E和mBFU-E集落数。在照射后第20天,可促进外周血WBC,RBC,Hct和Hb的恢复。说明骨髓基质细胞QXMSC1可促进辐射损伤小鼠造血功能恢复。此外,QXMSC1细胞条件培养上清液具有刺激体外CFU-GM集落生长的作用。A bone marrow stromal cell line QXMSCl was established by repeatedly passaging adhesive cells of murine marrow in vitro. The cell line was probably derived from macrophage by morphologic and functional identification. The model of hematopoietic injury was established by exposing BALB/c mice to 5 Gy γ-irradiation, and QXMSCl cells were infused into these mice. The proliferation of granulocyte/macrophage progenitors (CFU-GM) was observed by using semi-solid agar colony culture. The methylcellulose culture assay was used to measure the erythroid progenitors (CFU-E and mBFU-E). Peripheral blood parameters were measured by Coulter blood cell automatic counter. The results showed that bone marrow stromal cells can increase the number of nucleated bone marrow cells, CFU-GM, CFU-E and mBFU-E on the day 10 after irradiation. On the day 20, QXMSCl stromal cell can accelerate the recovery of WBC, RBC, Hct and Hb in peripheral blood. The results demonstrated that bone marrow stromal cell line QXMSCl can improve the recovery of the hematopoietic function of 5 Gy γ-irradiated mice. In addition, the conditioned medium of QXMSCl cells stimulated the growth of CFU-GM in vitro.
