摘 要:为了进一步阐明放射损伤复合烧伤(放烧复合伤)时骨髓造血基质细胞支持粒-巨噬系祖细胞造血能力的变化,采用小鼠骨髓基质细胞体外融合培养和粒-巨噬系祖细胞集落(CFU-GM)培养的方法,观察了5Gy照射复合15%Ⅲ度烧伤时基质细胞对支持CFU-GM生长的影响。结果表明:①单纯5Gy照射,单纯烧伤及放烧复合伤后,其骨髓基质细胞支持正常粒-巨噬系祖细胞造血的能力明显下降,以伤后3-5天最为显著,伤后10天仍未恢复至正常;②当骨髓基质细胞和粒-巨噬系祖细胞均受到致伤因素作用时,单纯照射及放烧复合伤组的CFU-GM形成能力下降更为明显,至正常对照组的10%以下,单纯烧伤组也有明显下降,但恢复较快;③在本研究所涉及的各种情况下,损伤的效应均有伤后3-5天达最低值的规律,之后可不同程度地恢复。实验结果提示,在上述损伤因素的作用下骨髓基质细胞的功能确已受到了不同程度的损伤,因此在治疗由此所引起的造血功能障碍时,同时加强促进基质细胞修复的措施,可能会取得更满意的疗效。The effect of marrow stromal cells on the growth of granulocyte-macrophage progenitor cells (CFU-GM) was investigated in mice with combined radiation-burn, radiation and burn injury. The method of semi-solid culture of CFU-GM over the confluent adherent stromal cell layers of marrow cells was adopted. The experimental data demonstrated that the numbers of normal marrow CFU-GM formed on the stromal cell layers from mice with combined radiation-burn, radiation and burn injury were all significantly reduced as compared with those of normal mice adherent layer at 1,3,5,7 and 10 days after injury. Whereas, under the same cultural condition, the CFU-GM in combined radiation-burn injury mice was decreased more severely, also was in the irradiated and burned mice marrow CFU-GM, however, the numbers of CFU-GM in burned animals were recovered and increased at day 7 and 10, respectively. Thus, the supporting function of bone marrow stromal cells on the growth of CFU-GM was impaired under the conditions of radiation, burn as well as conbined-burn injury in mice.
