摘  要:目的 :探讨性激素对子宫肌瘤细胞的作用机理。方法 :取活体子宫肌瘤组织进行细胞分离与原代培养至细胞汇合成片 ,分别加入不同浓度的雌二醇、睾酮 ,测定培养液中胰岛素样生长因子 -I(IGF -I)、胰岛素样生长因子结合蛋白 -3(IGFBP -3)的浓度 ,并与不加性激素组对照比较。结果 :对照组IGF -I、IGFBP -3分别为(344.25±52.18)μg/L和(9.22±0.33)μg/L ,雌二醇组为(292.25±15.65)μg/L和(8.28±0.46)μg/L ,睾酮组为(441.00±36.17)μg/L和(9.26±0.36)μg/L。与对照组相比 ,雌二醇抑制IGFBP -3的产生 (P<0.01) ;而睾酮则促进IGF -I的产生(P<0.01)。结论 :雌二醇抑制子宫肌瘤细胞产生IGFBP -3 ;而睾酮则促进IGF -I的产生 ,但对IGFBPObjective:To study the role of testosterone and estradiol on growth of human leimyoma cells.Methods:The cells derived from the living uteromyoma were isolated and cultured until they aggregated into flakes.The different concentrations of estradiol and testosterone were added respectively.The concentrations of IGF-I and IGFBP-3in the culture-media were measured.Results:IGF-1and IGFBP-3in the control were(344.25±52.18)μg/L and(9.22±0.33)μg/L respectively.IGF-1and IGFBP-3of the estradiol were(292.25±15.65)μg/L and(8.28±0.46)μg/L respectively.IGF-1and IGFBP-3of the testosterone were(441.00±36.17)μg/L and(9.26±0.36)μg/L respectively.Compared with the control,the productions of IGF-1and IGFBP-3in the cell culture media were suppressed by estradiol(P<0.05and P<0.01).The testosterone could increase IGF-I concentration in the cell media(P<0.01).Conclusion:Estradiol suppressed the production of IGFBP-3in uteromyoma cells.Testosterone promoted the production of IGF-I but did not play an evident role in the production of IGFBP-3.
