高温干燥对诱导伴性赤蚁蚕表达差异蛋白的影响

摘  要:以温敏性的伴性赤蚁蚕(Bombysmori)S伴为材料,用高温干燥(30℃、RH60%)与常温常湿(25℃、RH80%)的条件对蚕卵进行催青处理,测定了胚胎各发育阶段的可溶性蛋白含量,结果均发现催青前期变化不明显,而催青后期急剧下降,且在后期高温干燥区低于常温常湿区。另外,通过对这两个处理区的蚕卵可溶性蛋白进行SDS-PAGE筛选,寻找到孵化后期(胚胎发育到第8天时)的胚胎有明显差异蛋白带,并对这两条差异蛋白带进行扫描分析,发现一些蛋白含量发生变化;对这两个处理区较大差异蛋白区带进行分离提纯,经氨基酸组分测定表明,氨基酸的种类基本相同,而含量上存在差异,且氨基酸总含量常温常湿区低于高温干燥区。Sex-linked red ant silkworm (Bombys mori ) variety SBAN was used as material of temperature-sensitive silkworm breed. The breed was incubated under high temperature and low humidity (HTLH, 30 ℃, RH 60%) and normal temperature and humidity (NTNH, 25 ℃, RH80%) conditions, respectively. The soluble protein content was examined at each stage of embryonic development. It was found that the protein content of silkworm eggs had no obvious changes at the front stage, but decreased intensively at the later stage, and the protein content of embryo incubated under the HTLH condition was lower than that under the NTNH. Some differences were discovered from the bands of SBAN peptides under HTLH and NTNH conditions through SDS-PAGE selection when the eggs was incubated at the later stage. The peptides with differences were analyzed through scanning and some variation was found. When the protein bands with obvious difference were isolated and purified, the composition of amino acids had been determined. The results indicated that the types of amino acids were almost the same, but the quantity was different in content, and the total content of amino acids under HTLH condition was lower than that under NTNH.
