二期骨折愈合过程中骨痂软骨细胞凋亡与VEGF和TGF-β_1的表达

摘　　要：目的 :探讨二期骨折愈合过程中骨痂软骨细胞凋亡与VEGF和TGF - β1的表达变化。方法 :建立 16只SD大鼠股骨二期骨折愈合模型 ,分别于骨折后 1、2、3、4周取材 ,对骨痂进行HE染色及VEGF、TGF - β1免疫组化染色 ,同时用TUNEL法及琼脂糖凝胶电泳法检测细胞凋亡。结果 :VEGF骨折后第 1周在间充质细胞未见阳性表达 ,但软骨细胞、成骨细胞及新生网织骨小梁表面基质内呈强阳性表达 ;第 2、3周时软骨细胞中有强阳性表达 ;第 4周在软骨细胞中表达强度开始下降。TGF - β1第 1周在间充质细胞内呈弱表达 ;第 2周在软骨细胞中表达最强 ;第 3、4周其在软骨细胞中呈下降趋势。骨折愈合的不同时期都有细胞凋亡的发生 ,第 3周表现最为明显。结论 :在骨折愈合的不同时期 ,VEGF、TGF - β1表达的强弱有一定的差异。TGF - β1在骨折愈合早期可能起着促进间充质细胞向软骨细胞转化的作用 ,到第 2周其表达最强。VEGF在骨折的第 2周表达最强 ,到第 3、4周开始下降。软骨细胞的凋亡主要发生在骨折后的第 3周。Objective To study the expression change of VEGF , TGF-β 1 and apoptosis of chondrocytes during the second fracture healing. Methods An experimental model of the second fracture healing on rat's femur fracture was used to study the expression of VEGF and TGF-β 1 by means of immunohistochemistry ( ABC method ), HE staining results and the apoptosis of chondrocytes by TUNEL method and agarose gel eletrophoresis, using the samples taking from the different stages of healing (1,2,3,4 weeks after fracture). Results VEGF was strongly expressed in chondrocytes, osteoblasts and matrixes around bone trabecula of new woven bone in the 1st week after fracture.No expression of VEGF at the same time. 2nd and 3rd week after fracture ,VEGF was strongly expressed in chondrocytes. By the 4th week, the level of expression of VEGF started to decrease. TGF-β 1 was slightly expressed during the 1st week, its expression was the strongest in the 2nd week, and tend to decrease by the 3rd and 4th week. There were apoptosis of callus chondrocytes during the whole process of the second fracture healing. By the 3rd week, the phenomenon of apoptosis of callus chondrocytes was the strongest. Conclusion There were different expressions of VEGF,TGF-β 1 during different periods of the second fracture healing. TGF-β 1 can promote mesenchymal cell's transfer to chondrocyte during early stage of bone fracture healing, it was strongly expressed by the 2nd week.VEGF was strongly expressed during the 2nd week after bone fracture,its expression decreased by the 3rd and 4th week.Chondroctyes apoptosis happened mainly in the 3rd week after bone fracture. VEGF may directly take part in the process of chondrocyte apoptosis.
