摘  要:目的 观察蜂毒素对小鼠肝癌 H2 2 细胞增殖的影响 ,探讨其抗肿瘤的可能作用环节。方法 以 MTT法测定蜂毒素体外对小鼠肝癌 H2 2 细胞增殖的影响 ,运用流式细胞术分析增殖细胞核抗原 ( PCNA)的表达 ,并观察 iv蜂毒素对荷瘤小鼠的抑瘤作用。结果 蜂毒素对 H2 2 细胞的抑制作用随药物浓度增加而增强 ;蜂毒素作用于 H2 2 细胞后 ,细胞 PCNA平均荧光强度低于对照组 ;iv蜂毒素对小鼠皮下 H2 2 肝癌细胞具有明显的抑制作用 ,抑瘤作用随浓度增加而增强。结论 蜂毒素在体内外对小鼠肝癌 H2 2 细胞增殖均有明显的抑制作用 ,下调细胞Object To observe the effect of melittin on the growth and proliferation of H 22 hepatocarcinoma cells and study the possible mechanisms. Methods The proliferation inhibitory rate of H 22 cells treated with melittin was assayed by MTT. The volume of proliferating cell nuclear antigen (PCNA) expression of H 22 cells treated with melittin was analyzed with flow cytometer after being stained with immunocytochemical methods. The antitumor effect of melittin was observed in H 22 tumor burdened mice. Results The H 22 cell killing by melittin was dose dependent. After treatment with melittin, the fluorescence density of PCNA of H 22 cells decreased significantly. Melittin can inhibit the growth of H 22 hepatocarcinoma in vivo. Conclusion Melittin can inhibit the proliferation of H 22 cells both in vitro and in vivo. Down regulating the expression of PCNA of cells may be one of antitumor mechanisms.
