摘　　要：目的　研究小鼠肺气肿时 β 连环素 ( β catenin ,β cat)与广谱钙粘附素 (Pan cadherin ,Pan cad)在细支气管上皮的表达变化 ,探讨 β cat与肺气肿时小气道上皮损伤的关系。 方法　通过被动吸烟和经气管内注入木瓜蛋白酶两种方法建立肺气肿模型 ,采用弹力纤维染色观察肺泡壁弹力纤维变化 ;应用免疫组织化学方法 (SABC法 )和原位杂交技术研究 β cat、Pan cad、增殖细胞核抗原 (PCNA)以及 β catmRNA在细支气管上皮的表达。 结果　①两实验组细支气管上皮 β cat、β catmRNA、Pan cad在细胞膜表达降低 (均为P <0 0 1) ,两组间无明显差异 (P >0 0 5 )。其中 β cat在木瓜蛋白酶组和吸烟 6个月组分别有 3例 ( 37 5 % )和 4例 ( 5 0 % )在细胞浆表达 ,以及 2例 ( 2 5 % )与 3例 ( 37 5 % )细胞核表达。②两实验组细支气管上皮细胞核PCNA表达增多 (P <0 0 1)。结论　①肺气肿形成时 ,细支气管上皮主要呈增殖或过度增殖状态 ;②损伤因子 (如吸烟、木瓜蛋白酶等 )可引起细支气管上皮 β cat表达模式发生改变 ,即胞膜表达下调 ,胞质、胞核表达增加。Objective To study the expression of β catenin (β cat) in bronchiolar epithelium of the mice with emphysema induced by papain and smoking and evaluate the role of β cat and Pan cadherin (Pan cad) in the damage and repair of bronchiole. Methods By using HE staining and elastic fibers staining, morphological changes of lung were observed microscopically in the mice with emphysema induced by papain and smoking. The expression of β cat, Pan cad, proliferating cell nuclear antigen (PCNA) and β cat mRNA in bronchiolar epithelial cells was detected by immunohistochemistry and in situ hybridization. Results The expression levels of β cat, β cat mRNA and Pan cad of bronchiolar epithelium in the two experimental groups were lower than those in control group (all P <0 01). But the positive rate of PCNA expression in bronchiole epithelium was significantly higher in the experimental groups than those in control group ( P <0 01). The expression of β cat in the cytoplasmic and nuclei of bronchiole epithelium cells was detected in both smoking and papain induced models. Conclusion Bronchiole epithelium showed proliferation or excessive proliferation in emphysema. The injured factors as smoking, papain could result in the reduced expression of β cat in bronchiolar epithelium with up expression in the cytoplasmy and nuclei.
