脱细胞猪主动脉的组织结构及其免疫原性

摘　　要：目的:探讨猪主动脉(PA)、胰酶处理PA(TPA)、戊二醛处理PA(GPA)的结构及免疫原性。方法:无菌切取PA,部分经0.3%戊二醛处理0.5h,部分经胰酶处理72、96和120h;PA、GPA、TPA96(1×1cm2)分别埋植于A、B、C三组大鼠背部皮下,埋植第8、20和30天观察局部反应和血管片结构。反复冻融法分别制备PA、GPA和TPA96的组织细胞混合抗原,与完全弗氏佐剂、吐温-80混合制成复合乳剂抗原后免疫家兔,对流免疫电泳法测定兔血清抗体滴度。结果:TPA96与TPA72、TPA120相比,血管特性良好,细胞少,空隙多。TPA96、GPA复合乳剂抗原所致体液免疫反应相似,均弱于PA。A组大鼠第8天局部炎症反应剧烈,第20、30天渐轻。B组GPA呈黄褐色,仅第8天见明显炎症反应。C组大鼠各时点均未见明显炎症反应。第8、20天,血管片炎细胞浸润PA较严重,TPA96轻微,第30天各组间炎细胞浸润无统计学差异,但组织结构不同,TPA96可见多量细胞和小血管长入且血管组织与大鼠组织交织生长。结论:TPA96血管特性良好,免疫原性低,纤维可被逐渐代谢,细胞和血管易长入。Objective:To study the structure of porcine aortas treated with glutaraldehyde(GPA),trypsin(TPA)or nothing(PA)and their immunogenicity.Methods:Some PAs were treated with0.3%glutaraldehyde for0.5h,and others were acellularized by trypsin for72h,96h(TPA 96 )or120h separately in a5%CO 2 /95%air atmosphere at37℃under sterile condition.Vascular biological charac-teristics were studied.PA,GPA and TPA 96 patches(1×1cm 2 )were implanted subcutaneously on Wistar rats' back(group A,B and C correspondently),then were explanted and observed under light micro-scope8,20,and30days later.Rats' local response and patches structure were analyzed.Antigens,obtained from PA,GPA,TPA 96 separately by repeated freezing-melting process,were mixed with Com-plete Freund Adju vant and Tween-80to give emulsive antigens,which were used to immunize three rab-bits(1mg /kg antigen)correspondently.Finally,antibody concentration in those rabbits serum was mea-sured by counter immunoelectrophoresis.Results:TPA 96 had good vessel parameters,fewer cells and appropriate number of holes compared with TPA 72 and TPA 96 .Humoral immune reactions stimulated by GPA、TPA 96 emulsive antigen were similar but slighter than that by PA.In group A,local temperature rose in the8 th day and fell to normal on the20and30day,and evident edema and adherence were found consistently.When patches were explanted,slight bleeding occurred.In Group B,GPA became tan,with light edema and serious adherence to the surrounding tissue on the8th day.No edema andadherence were found in patches in Group C.On the8th and20th day,tissue inflammation was found the most serious in PA and the slightest in TPA 96 .On the30th day,inflammation in three groups ap-peared similar but their structure was different.TPA 96 grew interweavedly with rats' tissue and many cells and capillaries grew in it.Conclusion:TPA 96 has good vascular feature,with more holes and least immunogenicity.Vascular fibre can b
