摘  要:目的 研究银杏叶提取物对顺铂耳毒性的拮抗作用。方法 杂色健康豚鼠27只,随机分为生理盐水组、顺铂(CDDP)组、银杏叶提取物(Egb+CDDP)组,动态观察动物听阈变化及耳蜗外毛细胞形态学改变,并在损伤6天后,观察外毛细胞损伤率、耳蜗毛细胞凋亡情况。结果 CDDP组听阈及外毛细胞损伤率显著性升高,凋亡现象明显;Egb+CDDP组外毛细胞的损伤率及听阈损伤均明显减轻,仅出现早期凋亡表现:生理盐水组各项指标均无明显变化。结论 Egb对顺铂所致的耳毒性具有明显拮抗作用。Objective To investigate the antagonist effects of Ginkgo Biloba Extract (Egb) on the ototoxicity of cisplatin (CDDP) in guinea pigs. Methods Twenty-seven guinea pigs were randomly, divided into three groups normal saline, CDDP and Egb + CDDP groups. ABR thresholds at different time intervals were measured dynamicallyuntill that time significant changeing being observed. Then, Guinea pigs were decapitated for TEM and TUNEL determination to observe morphological changes of outer hair cells, injuring rate of hair cells and the apop-tosis of outer hair cells in the cochlea of animals. Results Hair cell loss, hearing threshold deteriorating and morphological changes of hair cells were very significantly severe, compared with that of the other two groups. Apoptotic cells could be seen after 6 days following cisplatin injection in the lower turn of tested cochlea. In contrast, no apoptotic cells were seen after 6 days following Egb used. Conclusion Egb holds protective effect on cochlea of Guinea pigs against ototoxicity of cisplation.
