水稻白叶枯病菌单抗杂交瘤细胞株的建立及应用研究

摘  要:经水稻白叶枯病原细菌(Xanthomonas campestris pv.oryzae)菌株Ks-6-6、Os-213、Yz-32,Yz-34,Yz-24'免疫的BALB/c小鼠脾细胞与骨髓瘤细胞SP2/0融合、筛选和克隆化,共获得12株稳定分泌该病原细菌单克隆抗体的杂交瘤细胞株。其抗体分属IgG3和IgG2b亚类。抗体与其它主要病原细菌的种或致病变种无交叉反应,能区分水稻白叶枯病原细菌3个不同血清型的菌株。腹水抗体效价在1:10~3—1:10~6左右。初步用于抗原决定簇的识别,能区分出6个抗原决定簇,并能将获得的63个菌株分为9个菌株组。By fusion of mouse myeloma cells (SP2/0-Ag14) and spleen cells derived from BALB/c mice immunized with the preparation of Xanthomonas campestris pv. oryzae Ks-6-6, Os-213, Yz-32, Yz-34 and Yz-24, 12 hybridoma cell lines secreting monoclonal antibodies were produced. All McAbs did not cross react with the other varieties of plant pathogenetic and non-pathogenetic bacteria. The McAbs could distinguish the variant serotypes of strains. Antibody titers of ascitis were about 1:10~3——1:10~6 when measured by ELISA method. The McAbs could differentiate 6 epitopes. Based on the epitopes, the 63 strains of X. campestris pv. oryzae were grouped into nine groups.
