双黄连注射液对内毒素诱导心肌损伤保护作用的研究

摘　　要：目的　观察双黄连注射液在内毒素诱导心肌损伤中的保护作用。方法　取SD雄性大鼠 18只随机分成正常对照组 (NC组 ,n =6 ) ,内毒素组 (ET组 ,n =6 ) ,双黄连组 (SHL组 ,n =6 ) ,在Langendorff装置上用Krebs Henseleit(KH)液对大鼠离体心脏行主动脉逆灌。在不同时点测定心功能指标 ,实验结束后取心肌组织测定一氧化氮 (NO)含量及琥珀酸脱氢酶 (SDH)、肌酸激酶 (CK)活性。结果　与ET组相比 ,SHL组在相应时点 ,左心室压差与心率乘积 (LVSP -LVEDP)×HR、LVDP、+dp/dtmax恢复百分率明显改善 ,对内毒素诱导的心肌SDH活性的降低、CK水平及NO含量的升高差异显著。结论　双黄连注射液具有明显的抗内毒素心肌损伤作用 ,其机制可能是通过降低心肌中NO的量 ,稳定心肌酶活性和能量代谢水平等途径 。Objective To observe effects of Shuanghuanglian Injection in myocardial injury induced by endotoxin. Methods 18 male SD rats,weighing(250±30g),were randomly divided into three groups:normal control group(NC,n=6),endotoxin group(ET,n=6) and Shuanghuanglian Injection group(SHL,n=6).The hearts of the rats were isolated and perfused Krebs-Henseleit(KH) (95%saturation95+5%CO 2) by using Langendorff apparatus at a constant pressure(8.33kPa)and temperature(37 ℃).The function of ventricular was measured at certain time(0 min,20 min,50 min,80 min).At the end of experiment,succinate dehydrogenase (SDH),creatine kinase (CK) and the concentration of nitric oxide (NO) in myocardial tissue were measured .Results Compared with ET group ,the percentage recovery of the product of left ventricular pressure difference and heart rate(LVSP-LVEDP)×HR, LVDP, +dp/dtmax were all increased apparently in SHL group . The activity of SDH ,CK and the concentration of NO were close to normal control group. Conclution Shuanghuanglian Injection has definite effect on anti-myocardial injury induced by endotoxin.It may be related to the decrease of the content level of NO, the stability of enzyme activities and the improvement of energy metabolism in myocardium.As a result, the resistance to myocardial injury induced by endotoxin will increase .
