摘  要:目的 观察通心络对血管成形术后家兔丝裂素活化蛋白激酶 (MAPK)表达的影响。方法 家兔 6 0只随机分为三组 (各组 2 0只 ) :A组 :对照组 ;B组 :高脂组 ;C组 :通心络组。采用高胆固醇喂养加两次动脉损伤的方法制作家兔血管成形术模型 ,再进行 4周的通心络治疗观察。测定家兔血脂指标 (TG、TC、LDL C、HDL C)、内皮素 (ET 1)和一氧化氮 (NO) ;HE染色测定管腔面积 (LA)、新生内膜面积 (NIA)和中膜面积 (MA)的变化 ;免疫组化染色定性观察MAPK的表达分布 ;用WesternBlot方法半定量测定MAPK的表达量。结果 通心络组血TC、LDL C低于高脂组 (P <0 .0 5 ) ;ET 1低于高脂组 ,NO高于高脂组 (P <0 .0 5 ) ;其管腔面积高于高脂组和对照组 (P <0 .0 5 ) ;新生内膜面积低于高脂组 ,高于对照组 (P <0 .0 5 )。MAPK阳性细胞面积明显低于高脂组 (P <0 .0 5 )。MAPK(ERK1和ERK2 )的蛋白表达量明显低于高脂组和对照组 (P <0 .0 5 )。结论 通心络通过抑制血管成形术后家兔MAPK的表达 。Objective To investigate the effect of Tong XinLuo and the mechanism of MAPK (mitogen activated protein kinases, ERK 1 and ERK 2) on the proliferation of neointimal after PTA (percutaneous translurminal angioplasty) model in atherosclerotic rabbit. Methods Set up atherosclerotic and PTA model in 60 rabbits, randomly divided into 3 groups,control group 20, high lipid group 20 and Tong XinLuo group 20. HE Staining and immunohistochemical analysis (ERK 1 and ERK 2) and Western Blot (ERK 1 and ERK 2) and blood biochemical tests (TG TC HDL C LDL C ET 1 NO) were done to evaluate the influence of MAPK on the proliferation of vascular intimal after PTA and the inhibition effect of Tong XinLuo. Using MET 1aMorph Image System to analysis all the materials acquired. Results It showed that Tong XinLuo group had larger lumen area (LA) and smaller neointimal area (NIA) than that of control group and high lipid group, Tong XinLuo group had lower level of serum TC LDL C and endothelin (ET 1) than high lipid group, but higher level of serum NO. Immunohistochemical analysis and Western Blot showed that the expressing of MAPK were increasing after PTA in high lipid group, but decreasing in Tong XinLuo group. Conclusion Tong XinLuo could inhibit the proliferation of intimal after PTA in atherosclerotic rabbit through inhibiting the expressing of MAPK.
