吡喹酮等药物口服与皮涂预防小鼠日本血吸虫感染的初步研究

摘　　要：目的 比较吡喹酮、氯硝柳胺、丙硫咪唑单独用药及联合用药灌胃预防小白鼠日本血吸虫感染及小剂量吡喹酮与两种透皮剂（二甲亚砜和Ａｚｏｎｅ）分别配成霜剂对小鼠透皮防护的效果。方法 灌胃给药实验分组为吡喹酮１２０ｍｇ／ｋｇ、吡喹酮 ８０ｍｇ／ｋｇ合并丙硫咪唑２０ｍｇ／ｋｇ、氯硝柳胺 １２０ｍｇ／ｋｇ、氯硝柳胺８０ｍｇ／ｋｇ合并丙硫咪唑２０ｍｇ／ｋｇ和空白对照五组，皮涂给药两个实验组和一个空白对照组，八组均为小鼠１２ｈ防护试验。结果 上述灌胃给药四组与对照组相比，减虫率分别为９１．７７％、９３．３２％、７６．３９％、４９．６９％；两种霜剂皮肤防护效呆均达１００％，且各组减虫率与抗体阳性率相一致。结论 灌胃给药各组均有明显的防护作用，且低剂量联合用药有一定的增效作用；同时进一步证实吡喹酮霜剂防护效果确实，两种透皮吸收促进剂作用明显，为血吸虫现场防护提供了可靠依据。Objective To compare the protective effects of intragastrical single and joint administration of praziquantel, niclosamide and albendazole against mice’s infection with Schistosoma japonicum, and the protective effects of praziquantel creams including different dennal penetration enhancers. Methods Four groups for oral ad- ministration: 120mg/kg praziquantel, 80mg/kg pra.ziquantel and 20mg/kg albendazole, 120mg/kg niclosarnide, 80mg/kg niclosamide and 20mg/kg albendazole, an oral control group. Two groups for dennal administration: praz- iquantel cream with azone, that with dimethyl sulfoxide, a dermal control group. All for 12h protective test on mice. Results In comparison with the oral contra1 group, the reduction rates of adult womis of four oral adminis- trational groups were 91 .77%, 93 . 32%, 76. 39% and 49. 69%, respectively. Tin comparison with the dernial control group, that of two dermal administrational groups both were 100%, and that of all groups were unanimous with their antibody positive rates. Conclusion The intragastrical administrational groups showed significant pm- tective effects, and the oral joint administrational groups appeared certain increasing effects. Meanwhile, the der- mal protective test of mice further proved the effect of praziquantel cream true, azone and dimethyl sulfoxide both were veiy effective cutaneous penetration enhancers.
