摘 要:应用 TGGE指纹图谱技术对两个曝气池细菌种群的动态变化及多样性进行了研究。每 3d进行 1次 ,共 8个监测时期中同一曝气池活性污泥的 1 6Sr DNA V3- PCR TGGE指纹图谱基本一致 ,图谱间的相似性系数 (Cs)为 1 0 0 %。同一曝气池不同位点活性污泥的 TGGE指纹图谱也完全一致。功能不同曝气池活性污泥 TGGE指纹图谱存在差异 ,Cs为 83.3%。对 TJ1活性污泥 TGGE图谱中 9条主要条带回收、扩增、克隆建库 ,每个条带选 4个转化子进行序列分析 ,结果显示 TGGE条带是由序列不同的片段组成。 32个序列在97%的相似性下分成 1 6个分类操作单元 (OTU) ,1 4个 OTU与 Gen Bank中已登录的细菌种群的同源性≥ 9 7% ,2个 OTU的同源性为 95%和 94%。与 1 0个 OTU同源性较高的细菌类群是在活性污泥或污染环境分离或发现的 ,与 8个 OTU相似的细菌类群目前尚无法分离培养。TGGE analysis of 16S rDNA gene V3 region was used to study the population dynami cs and bacterial diversity in two functionally different aeration tanks (TJ1 and TJ2) within a phenol-degrading activated sludge process. The band patterns pro duced from samples collected at 3-day intervals over 8 time points from each ae ration tank demonstrated a high degree of similarity, with Cs values at 100 %. TGGE fingerprinting for activated sludge samples from different sites in one tank showed identical patterns, but that from functionally different tank exhibi ted both common bands and unique bands, with a Cs value of 83.3%. Nine major bands from TGGE profiles of TJ1 sample was eluted from gel, reamplified, cloned and a library established. Four clones from each band were sequenced and more t han one sequence type was found in each band demonstrating the co-migration of DNA fragments with substantial sequence variation in TGGE bands. The 32 sequence s for which there was no indication of chimerism were aligned with the sequences of closest relatives in GenBank. The results suggested that they belonged to 16 OTUs at 97% homology cutoff. 14 OTUs shared 97%~100% homology with known seque nces in the database while 2 OTUs had 94%~95% homology with known sequences. Am ong all the OTUs, 10 were similar to bacteria in database that were reported to have been isolated from different activated sludge systems and contaminated envi ronments. The isolates from 8 OTUs grouped together with some uncultured eubacte ria in the database. A phylogenetic analysis of 16 OTUs classified them into two major groups: the OTU13 lineage and the other 15 OTUs lineage, the later was cl assified further into 4 subgroups. The sequence diversity from TJ1 revealed that high diversity of bacterial populations existed in the industrial phenol biorem ediation system and the community structure was relatively stable over time.
