摘 要:目的 :探讨败血症大鼠不同时间血浆促炎症细胞因子 (IL - 8、TNF -α)、外周血液和腹腔渗出液中炎性细胞的变化规律以及山莨菪碱 (6 5 4 - 2 )干预的影响。方法 :采用盲肠结扎加穿刺法复制大鼠败血症模型 ,分别在 0h、8h、16h取血和腹腔渗出液 ,采用酶联免疫吸附试验 (ELISA法 )测定血浆IL - 8、TNF -α含量 ;常规进行外周血和腹腔渗出液中白细胞计数。结果 :败血症组和 6 5 4 - 2干预组 8h、16h血浆IL - 8、TNF -α水平均较 0h明显增高 (P <0 .0 1) ;败血症组大鼠在 8h、16h血浆IL - 8、TNF -α均高于假手术对照组 (P <0 .0 5 ) ;6 5 4 - 2干预组上述 2种细胞因子水平在 8h、16h均低于败血症组 (P <0 .0 5 )。败血症组和 6 5 4 - 2干预组外周血白细胞计数 8h、16h均较 0h明显降低 (P <0 .0 1或 0 .0 5 ) ,且均明显低于假手术对照组 ;而败血症组和 6 5 4 - 2干预组大鼠腹腔渗出液中白细胞计数 8h、16h均较 0h显著增高 (P <0 .0 1) ,且均明显高于假手术对照组。结论 :IL - 8、TNF -α在大鼠败血症发生发展过程中可能起重要作用 ;败血症大鼠外周血和腹腔渗出液中炎性细胞的异常分布现象可能与IL - 8、TNF -α有关。 6 5 4 - 2可能有抑制IL - 8、TNF -α过度产生或释放作用。Objective: To detect the changes of proinflammatory cytokines (IL-8,TNF-α)in plasma and inflammatory cells in peripheral blood and abdominal effusion in different times and anisodamine(654-2) interference in septicemic rats.Methods: The sepsis models were replicated by means of cecum ligature-puncture. The levels of IL-8 & TNF-α were measured through enzyme linked immunosorbent assay(ELISA) and the numbers of leucocytes in peripheral blood and abdominal effusion were counted down with routine method in 0,8 and 16 hours postexperimentally. Results: In both sepsis and 654-2 interference group ,the levels of plasma IL-8 & TNF-α in 8 and 16 hours after the experiments were noticeably higher than those in 0 hour( P <0.01) ,and IL-8 and TNF-α levels of sepsis group in 8 and 16 hours were obviously higher than those of control group with sham operations ( P <0.05).But levels of IL-8 & TNF-α in 654-2 interference group were decreased sepsis group in 8 and 16 hours after the experiments( P <0.05). The numbers of leukocyte in peripheral blood of septicemic and 654-2 interference group in 8 and 16 hours postexperimentally were evidently reduced than those in 0 hour ( P <0.01 or 0.05) and were all decreased to contrast group with sham operations . The leukocyte numbers of abdominal effusions in 8 and 16 hours were clearly increased to those of 0 hour in sepsis group and 654-2 interference group ( P <0.01) and were all increased to contrast group with sham operations. Conclusion: IL-8 and TNF-α may play an important role in the course of rats'sepsis and the abnormal distribution of inflammatory cells of sepsis in rats were closely related to IL-8 and TNF-α.Anisodamine may inhibit the overproducing or discharging of IL-8 and TNF-α.
