侵染云南白肋烟的中国番茄黄化曲叶病毒及伴随卫星DNA分子的基因组特征

摘　　要：从中国云南省大理地区表现曲叶症状的白肋烟(Nicotinatabacum WhiteBurley)上分离到病毒分离物Y43,该病毒可经烟粉虱(Bemisiatabaci)及嫁接传播。用15种粉虱传双生病毒的单抗对病样进行TAS-ELISA检测,结果表明,该病毒属菜豆金色花叶病毒属(Begomovirus)病毒。对DNA-A1.7kb基因组序列测定和分析表明,Y43与中国广西报道的中国番茄黄化曲叶病毒(TomatoyellowleafcurlChinavirus,TYLCCNV)同源性达89%,其中外壳蛋白(CP)氨基酸同源性达96%,因此Y43应为TYLCCNV的一个新分离物。进一步研究发现,Y43还伴随着一个长1349nt的卫星DNA分子(DNAβ)。Y43DNAβ与AYVVDNAβ、BYVMVDNAβ和CLCuVDNAβ的同源性较低,而与中国分离的烟草曲叶病毒Y5DNAβ和Y8DNAβ的亲缘关系较近。DNAβ可能编码7个分子量超过3.5kD的ORF,其中C1推测为有功能的ORF。The virus isolate Y43 was collected from Nicotiana tabacum White Burley plant showing leaf curl symptom in Dali, Yunnan Province. Y43 can be transmitted to tobacco plants by whitefly (Bemisia tabaci ) and grafting. The reaction with 15 monoclonal antibodies raised against particles of whitefly-transmitted geminivirus demonstrated that the virus isolate was a begomovirus . The analysis of 1.7 kb DNA-A sequences of Y43 revealed that it shared 89% nucleotide sequence identity with Tomato yellow leaf curl China virus (TYLCCNV) DNA-A, while 96% amino acid sequence identity was found for coat protein(CP) gene. Therefore, Y43 was taxonomically considered as an new isolate of TYLCCNV. Further studies showed that Y43 was associated with a novel satellite DNA molecule (DNAβ). Sequence analysis revealed that Y43 DNAβcontained 1 349 nt and had capacity to code seven possible ORFs with molecule weight over 3.5 kD, among which C1 (13.8 kD) was proposed to be functional. Alignment of published DNAβsequences showed that Y43 DNAβwas most related to Y5 and Y8 DNAβ, associated with Tomato yellow leaf curl China virus , and was distant from AYVV, BYVMV and CLCuV DNAβ.
