摘  要:目的 应用鹅膏蕈氨酸 (IBA)和 D-半乳糖 (D- gal)诱发 Wistar大鼠认知障碍模型 ,观察补充外源性雌激素的治疗作用与机理。方法  1认知障碍组 :应用 IBA(1μl)一次性微量注入大鼠 Meynert基底核 ,同时大鼠皮下注射 D- gal(48mg/ Kg) ,制作模型。 2治疗组 :按模型组方法制备模型 ,在应用 IBA后 7d开始应用苯甲酸雌二醇 (1 mg/ Kg)和黄体酮 (1 0 mg/ Kg)治疗 ,每 7d皮下注射 1次 ,治疗时间 1 50 d。3假手术组 :健康大鼠按模型制作过程操作 ,应用生理盐水替代 IBA和 D- gal,注射用水替代雌二醇和黄体酮皮下注射。用 MG- 2迷宫观察大鼠至其测试达到连续 1 0次中有 9次正确反应所需要的电击次数 ,作为学习记忆成绩。HE染色和 Bielschowsky染色观察脑组织变化。结果 应用 IBA和 D- gal1 50 d后 ,模型组大鼠学习记忆能力较其他组明显低下 ,其电击次数 (2 5.5± 1 2 .39次 )明显高于治疗组 (3.80± 3.83次 )和假手术组 (2 .3± 4.50次 ) ,均 P<0 .0 5;而治疗组和假手术组比较无显著性差异 (P>0 .0 5)。光镜下海马皮层平均单个视野中锥体细胞计数模型组 (3.2 5± 2 .60个 )明显少于治疗组 (1 5.0± 6.35个 )和假手术组 (1 7.85± 1 1 .1 9个 ) ,均 P<0 .0 5;而治疗组和假手术组无显著性差异 (P>0 .0 5)。Objective The model of rat as cognition disorder was established by using Ibotenic Acid(IBA) and D-galactose(D-gal), and the effect and mechanism of the treatment with estrogen supplement theray were observed.Methods ①The model of Cognition disorder group(model group): The model was made by the injection of IBA(1 μl) into the Meynert nucler using microdrive injector only one time. D-gal(48 mg/Kg) was also used through subcutaneous injection.②The treatment group: The same methods to make the disease were used as the model group. But the rats was treated with the estrogen supplement therapy(estrogen 1mg/Kg). ③Sham-operated group: the healthy rats were used without any drugs. The standard that rats arrived at 9/10 correct times in the MG-2 maze were regarded as the ability of learning and memory. The histopathological changes of brain were observed according to HE and Bielschowsky dyeing.Results IBA and D-gal can cause damage of rats in learning and memory. After the injection 150 days, the times of learning and memory of the model group(25.5±12.39 times) was much higher than the treatment group(3.80±3.83 times) and the sham-operated group(2.3±4.50 times), P<0.05,0.05. But the difference of the treatment group and the sham-operated group was not distinct. The average amounts of pyramidal cells in one visual field were obviously decreased in the model group(3.25±2.60) compared with the treatment group(15.0±6.35) and the sham-operated group(17.85±11.19) in CA1 of hippocampus, P<0.05,0.05. But the treatment group had no obviously difference with the sham-operated group. The NFT and SP were not observed in every group.Conclusions IBA and D-gal can result in cognition disorder in rates, and the estrogen supplement therapy had efficiency in the treatment of the disease.
