摘 要:目的 从一氧化氮 (NO)和肿瘤坏死因子 (TNF)的变化探讨大黄及其提取物抗脑缺血损伤的机制。方法 采用双侧颈总动脉缺血模型 ,分为假手术组、模型组、尼莫地平组、大黄组、大黄部位 1组、大黄部位 2组、大黄部位 3组、大黄部位 4组。观察脑组织病理改变 ,测定脑电图、脑组织和血清 NO和 TNF- α。结果 模型组脑电图幅值降低、脑组织病理损伤严重 ,血清和脑组织中 NO、TNF- α水平增高。与模型组比较 ,尼莫地平组和其他用药组脑组织病理损伤减轻 ;除了大黄部位 2组外 ,其他用药各组脑电幅值增高 ;尼莫地平组、大黄组、大黄部位 3组、大黄部位 4组血清和脑组织 NO、TNF- α水平显著降低 ;大黄部位 1组脑组织 TNF- α水平和血清 NO、TNF- α水平降低显著 ,大黄部位 2组脑组织 NO含量和血清、脑组织TNF-α水平皆降低明显。结论 大黄及 4个不同部位可改善缺血后脑组织损伤 ,其保护机制可能与减轻 NO、TNF介导的神经毒性作用有关。on Nitric Oxide (NO) and Tumor Necrosis Factor (TNF).Methods Applying the model of bilateral common carotid artery ischemia . SD rats were divided randomly into sham-operated group, model group, Nimodipine(ND) group, Rhubarb(R) group, Rhubarb 1 ( R1)group, Rhubarb 2 group ( R 2), Rhubarb 3 (R3) group and Rhubarb 4 (R4) group. The brain pathological variation was observed and the contents of brain and serum NO and TNF-α were determined and EEG was recorded.Results The amplitude of EEG was lower, the cerebral pathological injury was more serious, the contents of brain and serum NO and TNF-α were higher in the model group than those of the sham-operated group,Compared with the model group, cerebral pathological injury was alleviated in all the group with drug,the amplitude of EEG was higher in other groups that were treated with drug but the R 2's. The contents of brain and serum Non and TNF-α significintly decreased in the ND's, the R's,the R3's and the R4's, the level of brain and serum NO and TNF-α, serum TNF was marketly lower in the R1's,the level of brain and serum TNF-α and brain NO was obviously decreased in the R2's.Conclusions Rhubarb and its four different parts, especially the R3 and the R4,could alleviate cerebral ischemia injury, the effect mechanism might related to inhibite the injury induced by NO and TNF.
