摘  要:目的合成α-D-葡萄糖苷酶检测底物对硝基苯基α-D-葡萄糖苷,建立临床检测方法。 方法采用Westphal-Feier合成法制备对硝基苯基α-D-葡萄糖苷;依据α-葡萄糖苷酶作用于底物的动力学性质,建立α-葡萄糖苷酶的检验方法。 结果获得高纯度底物对硝基苯基α-D-葡萄糖苷,纯度>99.0%,并开发成试剂盒,批内变异<2.5%,批间变异<3.8%,阳检率85.7%。 结论试剂盒符合临床应用要求。Objective To synthesize p - nitrophenyl -α-D-glucopyranoside as a substrate for α- glucosidase, and develope a method for testing its activity. Methods The substrate was prepared by Westphal and Feier synthesis. A quantitative method was established according to α-glucosidase' s kinetic characters on the substrate. Results A highly purified substrate (purity >99.0% ) was obtained. A kit for α-glucosidase was made with OCV < 2. 5% , RCV < 3. 8% , and sensitivity 85. 7% . Conclusion The kit can be reliably used in clinics.
