糖尿病大鼠肾组织中NF-κB和MMP-9及TIMP-1表达关系的研究

摘　　要：目的 :研究糖尿病大鼠肾组织中核因子 κB(NF κB)和基质金属蛋白酶 9(MMP 9)及组织基质蛋白酶抑制因子 1(TIMP 1)表达之间的关系方法 :将 3 0只雄性Wistar大鼠随机分为正常对照组 (C组 )、糖尿病未治疗组 (D组 )和糖尿病苯那普利治疗组 (B组 ) ,每组10只 ,利用链脲佐菌素 (STZ)诱导糖尿病模型 ,应用免疫组化方法研究大鼠肾组织中NF κB、MMP 9和TIMP 1的表达 ,并利用HPIAS 10 0 0型医学彩色图像分析系统进行图像分析。结果 :各组间的差异均有显著性意义 (P <0 .0 1)。NF κB与MMP 9成明显负相关 (r =-0 .882 67,P <0 .0 1) ,NF κB与TIMP 1无明显相关 (r =0 .5 2 981,P >0 .0 5 ) ,NF κB与MMP 9/TIMP 1的比值亦成明显负相关 (r =-0 .8685 0 ,P <0 .0 1)。结论 :NF κB对糖尿病大鼠肾组织中MMP 9的表达起重要作用 。Objective: To investigate the relation of expression for the nuclear factor κB(NF κB)?matrix metalloproteinase 9(MMP 9) and tissue inhibitor of metalloproteinase 1(TIMP 1) in kidney tissue of diabetes rats.Method: Thirty male wistar rats were divided into 3 groups randomly as follows the normal control group(C group)?diabetes mellitus group(D group) and treatment by benazepril on diabetes mellitus rats group(B group),10 in each group.Diabetes was induced by administrating streptozotocin(STZ).The expressions of NF κB,MMP 9 and TIMP 1 in kidney tissue were studied by immunohistochemistry(SP) method.The results were analyzed by the type of HPIAS 1000 medical colour picture analysis system.Results: The results were difference among three groups was significant( P <0.01)?The expression of NF κB and MMP 9 took on negative correlation significantly( r=-0.88267,P<0.01 ).The expression of NF κB and TIMP 1 took no significant correlation( r=0.52981,P>0.05 ).The expression of NF κB and the ratio fo MMP 9/TIMP 1 also took on negative correlation significantly( r=-0.86850,P<0.01 ).Conclusion: NF κB takes an important role on the expression of MMP 9 in kidney tissue of diabetes rats,and this kind of mechanism may take part in the occurrence and progression of diabetic nephropathy.
