反义bcl-2-正义bcl-Xs逆转录病毒重组体的构建

摘　　要：构建一种反义抗细胞凋亡基因-正义促细胞凋亡基因相连结的新型表达质粒,以探讨其对细胞凋亡的影响。用分子克隆方法,成功地构建了反义bcl-2-正义bcl-Xs逆转录病毒重组体,初步结果显示此重组体比反义bcl-2或正义bcl-Xs逆转录病毒重组体具有更强的促细胞凋亡作用。该新型表达质粒的构建为探讨细胞凋亡提供了新工具,新思路。A new kind of expression vector which contains conjoined antisense antiapoptosic gene and sense apoptosic gene was constructed for studying its effects on apoptosis. To construct this kind of expression vector, method of molecular cloning was used. Results showed that the retroviral recombinants containing conjoined antisense bcl-2 and sense bcl-Xs has a stronger synergetic effects on the apoptosis of Jurkat celis than the retroviral recombinants containing antisense bcl-2 or sense bcl-Xs. Conclusion suggested that the new kind of expression vector could serve as a new tools and methods for studying apoptosis.
